CP-25 combined with MTX/ LEF ameliorates the progression of adjuvant-induced arthritis by the inhibition on GRK2 translocation
ConclusionEquivalent therapeutic effects were observed between CP-25-combination groups and high-dose MTX/LEF groups. CP-25 inhibited p-ERK by reducing the membrane expression of GRK2 in FLS from AA rats.Graphical abstractFigure. Mechanism of CP-25 combined with MTX/LEF to ameliorate AA ratsIn the one hand, CP-25 alone and in combination blocked the activation of P38, ERK, and JNK. CP-25 combined with MTX/LEF exerts synergistic effects to inhibit inflammation of AA rats by inhibiting the MAPK pathway. In the other hand, CP-25 inhibited GRK2 translocation in AA rats which further restored the inhibition of GRK2 on p-ERK. CP...

Publication date: January 2019Source: Biomedicine & Pharmacotherapy, Volume 109Author(s): Zhenhuan Li, Furong Wu, Dafeng Xu, Zhongzheng Zhi, Guanghui XuAbstractSpinal cord injury (SCI) is a devastating event, leading to the progression of chronic neuropathic pain syndrome. Triggering receptor expressed on myeloid cells 1 (TREM1) is an innate immune receptor expressed on neutrophils and monocytes/macrophages. TREM1 enhances inflammatory response in various models of diseases, but its significance in SCI remains unclear. In the present study, we attempted to explore the effects of TREM1 on the regulation of SCI. Spinal cord ...

Down-regulation of JMJD5 suppresses metastasis and induces apoptosis in oral squamous cell carcinoma by regulating p53/NF-κB pathway
Publication date: January 2019Source: Biomedicine & Pharmacotherapy, Volume 109Author(s): Yuan Yao, Wen-Ying Zhou, Rui-Xin HeAbstractThe prognosis of oral squamous cell carcinoma (OSCC) patients remains unclear, and a better understanding of the underlying molecular mechanisms is urgently required. Jumonji-C (JmjC) domain-containing protein 5 (JMJD5), renamed KDM8, has been implicated in tumorigenesis, circadian rhythm modulation, embryological development, and osteoclastogenesis. In the present study, we found that JMJD5 was over-expressed in patients with OSCC by real-time quantitative PCR (qPCR), western blot and immuno...

Cytokine augments the sorafenib-induced apoptosis in Huh7 liver cancer cell by inducing mitochondrial fragmentation and activating MAPK-JNK signalling pathway
Publication date: February 2019Source: Biomedicine & Pharmacotherapy, Volume 110Author(s): Lijuan Zhang, Shuping Li, Rong Wang, Changyuan Chen, Wen Ma, Hongyi CaiAbstractSorafenib is a standard targeted drug used to treat hepatocellular carcinoma (HCC). Notably, cytokine has been found to further enhance the therapeutic effectiveness of the targeted drug. Thereby, the aim of this study is to verify whether cytokine IL-2 could increase the anti-cancer effects of sorafenib on liver cancer in vitro. Huh7 cells were used in the present study and the cell apoptosis and migration were determined in response to sorafenib treatmen...

Effect of miR-30e regulating NK cell activities on immune tolerance of maternal-fetal interface by targeting PRF1
Conclusion: Up-regulated miR-30e can reduce the cytotoxicity of PB-NK cells and D-NK cells by targeting PRF1, whereby inhibiting Th1 tolerance phenotype and inducing Th2 immunodominance. miR-30e may be contributive to creating a micro-immune tolerance environment of maternal-fetal interface.Graphical abstract

Effects of EIF3B gene downregulation on apoptosis and proliferation of human ovarian cancer SKOV3 and HO-8910 cells
Publication date: January 2019Source: Biomedicine & Pharmacotherapy, Volume 109Author(s): Lei Wang, Ling OuyangAbstractIn order to investigate the role of EIF3B (eukaryotic translation initiation factor 3B, EIF3B) gene in the proliferation and apoptosis of ovarian cancer cells, a lentiviral vector system and shEIF3B lentiviral vector were constructed to transfect human ovarian cancer cells. SKOV3 and HO-8910 cells were used in this experiment. The cell growth was detected by Celigo cell counting assay, the apoptosis rate was measured by flow cytometry and the cell proliferation ability of lentivirus transfected cells was t...

Baicalein stimulates fibroblast growth factor 21 expression by up-regulating retinoic acid receptor-related orphan receptor α in C2C12 myotubes
Publication date: January 2019Source: Biomedicine & Pharmacotherapy, Volume 109Author(s): Takao Hirai, Kohei Nomura, Rie Ikai, Ken-ichi Nakashima, Makoto InoueAbstractRetinoic acid receptor-related orphan receptor (ROR) α has been implicated in various physiological functions, including the immune system, inflammation, and circadian rhythms. In the present study, the synthetic RORα/γ agonist SR1078 stimulated the production and gene expression of fibroblast growth factor 21 (FGF21) in C2C12 myotubes. FGF21, a member of the FGF family, plays important roles in the regulation of peripheral glucose tolerance and lipid meta...

Activation of TGR5 promotes osteoblastic cell differentiation and mineralization
Publication date: December 2018Source: Biomedicine & Pharmacotherapy, Volume 108Author(s): Qingfeng Wang, Guoqiang Wang, Bin Wang, Huilin YangAbstractImpairment of normal osteoblast differentiation has been associated with bone loss-related disorders, such as osteoporosis. Takeda G-protein coupled receptor 5 (TGR5) has been identified as an important modulator of bile acid and energy homeostasis. Little information regarding the effects of TGR5 on osteoblastic bone formation and matrix mineralization has been reported before. In the current study, we found that TGR5 was expressed in osteoblast-like cell line MC3T3-E1 cells...

Oxidative stress-elicited YY1 potentiates antioxidative response via enhancement of NRF2-driven transcriptional activity: A potential neuronal defensive mechanism against ischemia/reperfusion cerebral injury
Publication date: December 2018Source: Biomedicine & Pharmacotherapy, Volume 108Author(s): Wei Liu, Qingdong Guo, Haikang ZhaoAbstractCerebral ischemia challenge evokes an adaptive defensive system through the induction of antioxidant enzymes. Many of such antioxidant enzymes are regulated essentially by the nuclear factor erythroid 2-related factor-2 (NRF2). Compelling evidence supports that targeting NRF2 signaling is a promising therapeutic strategy to alleviate post-I/R brain injury, but the molecular mechanisms underlying this strategy have yet to be elucidated. Herein, we show that the expression of transcription fac...
