SiRNA-Mediated Flotillin-2 (Flot2) Downregulation Inhibits Cell Proliferation, Migration, and Invasion in Gastric Carcinoma Cells.
In conclusion, the present study suggests that the Flot2 protein expression is significantly correlated with cancer progression and poor prognosis in gastric carcinomas, probably due to its role in the regulation of cell proliferation, migration, and invasion in gastric carcinoma cells. Enhancement of Drug Sensitivity by Knockdown of HIF-1α in Gastric Carcinoma Cells.
In this study, the effects of hypoxia-inducible factor-1α (HIF-1α) on gastric carcinoma (GC) drug resistance through apoptosis-related genes are investigated. First, HIF-1α-specific siRNA was synthetized and transfected into drug-resistant GC cell line OCUM-2MD3/L-OHP. Then MTT assay was applied to test the inhibition rate of GC cells by 5-fluorouracil (5-FU) and oxaliplatin (L-OHP). After that, flow cytometry (FCM) was applied to measure apoptosis rate. qPCR and Western blot assay were employed to detect HIF-1α and apoptosis-related genes. Results showed that HIF-1α in OCUM-2MD3/L-OHP cells was higher than that in OC...

Enhancement of Chemosensitivity by Stathmin-1 Silencing in Gastric Cancer Cells In Situ and In Vivo.
Authors: Meng ZJ, Tao K Abstract Reports show that the stathmin gene may have a close relationship with tumor chemotherapeutic sensitivity. However, the effect of stathmin-1 on the chemosensitivity of gastric cancer to docetaxel has not been clearly determined. siRNA targeting stathmin-1 was introduced. The cell growth inhibition, expression of associated proteins, cell cycle, and apoptosis were evaluated by MTT, Western blot, and flow cytometric assays, respectively. The influence of silencing stathmin-1 was detected in situ and in vivo. SGC7901/docetaxel cells are the drug-resistant cells. After silencing stathmi...

Gastrin enhances autophagy and promotes gastric carcinoma proliferation via inducing AMPK α.
In conclusion, these findings indicate that gastrin plays a tumorigenic role by promoting autophagy in gastric cancer, and may provide a novel therapeutic target for gastric cancer treatment. PMID: 28059052 [PubMed - as supplied by publisher]

CSTB Downregulation Promotes Cell Proliferation and Migration and Suppresses Apoptosis in Gastric Cancer SGC-7901 Cell Line.
This study aimed to investigate the pivotal role of cystatin B (CSTB) in the development of gastric cancer and to explore its possible regulatory mechanism. Human gastric cancer SGC-7901 cells as a model in vitro were transfected with plasmid PCDNA3.1-CSTB and siRNA-CSTB using Lipofectamine 2000. Quantitative real-time PCR (qRT-PCR) and Western blotting were performed to determine the relative expression of CSTB and PI3K/Akt/mTOR pathway-related protein. Moreover, MTT assay, Transwell assay, and flow cytometry were used to assess cell proliferation, migration, and apoptosis, respectively. The results showed that CSTB was s...

DKK3 Overexpression Increases Malignant Properties of Head and Neck Squamous Cell Carcinoma Cells.
Authors: Katase N, Nishimatsu SI, Yamauchi A, Yamamura M, Terada K, Itadani M, Okada N, Hassan NMM, Nagatsuka H, Ikeda T, Nohno T, Fujita S Abstract DKK3, a member of dickkopf WNT signaling pathway inhibitor family, is believed to be a tumor suppressor, because of itsreduced expression in cancer cells. However, our previous studies have revealed that DKK3 expression is predominantly observed in head and neck, and oral squamous cell carcinoma (HNSCC/OSCC). Interestingly, HNSCC/OSCC patients with DKK3 expression showed high metastasis rate and poorer survival, and siRNA-mediated knockdown of DKK3 in HNSCC-derived can...

miR-203 inhibits the invasion and EMT of gastric cancer cells by directly targeting Annexin A4.
Authors: Li J, Zhang B, Cui J, Liang Z, Liu K Abstract Many studies have shown that down-regulated miR-203 level is in a variety of cancers including gastric cancer (GC). However, the precise molecule mechanisms of miR-203 in GC have not been well clarified. In current study, we investigated the biological functions and molecular mechanisms of miR-203 in GC cell lines. We found that miR-203 is down-regulated in GC tissues and cell lines. Moreover, the low level of miR-203 was associated with increased expression of Annexin A4 in GC tissues and cell lines. The invasion and EMT of GC cells was suppressed by overexpre...
