Two new naphthalenones from Diospyros undulata stem bark and their cytotoxic activity

Suchaichit, Nattawut Suchaichit, Kwanjai Kanokmedhakul, Kitisak Poopasit, Panawan Moosophon, Somdej Kanokmedhakul Two new naphthalenones, cis-isoshinanolone-4-acetate (1) and 3,4-dihydro-4β-hydroxy-5,6-dimethoxy-2α-methyl-1(2H)-naphthalenone (2), were isolated together with three known naphthoquinone derivatives (3-5), four dimeric naphthoquinones (6-9), two coumarins (10-11) and four triterpenoids (12-15) from the stem bark of Diospyros undulata. Their structures were characterized by intensive spectroscopic data (1D and 2D NMR, IR, UV and MS). Compounds 4, 7 and 9 exhibited strong cytotoxicity against three cancer cell lines including KB, MCF-7 and NCI-H187 with IC50 values in the range of 0.35 to 9.12 μM. Naphthoquinone 5 demonstrated moderate cytotoxicity against MCF-7 (IC50 26.05 μM) without cytotoxicity to Vero cells. Graphical abstract
