TGF-β Targets the Hippo Pathway Scaffold RASSF1A to Facilitate YAP/SMAD2 Nuclear Translocation

Pefani et al. identify that the tumor suppressor RASSF1A is an integral component in TGF-β signaling. TGF-β stimulates RASSF1A recruitment to TGF-βRI at the cell membrane, where it is targeted for degradation by the E3 ligase ITCH. Degradation of RASSF1A allows YAP/SMAD interaction, nuclear localization, and TGF-β−mediated transcription.
