A new method of estimating prevalence of childhood cancer survivors (POCCS): example of the 20-year prevalence in The Netherlands
CONCLUSIONS: Using the POCCS model and aggregated cancer data, our estimates of childhood cancer survivors limited-duration prevalence were consistent with those computed by a standard method requiring individual cancer records. The POCCS method provides relevant information for planning follow-up and care for childhood cancer survivors.PMID:37738448 | DOI:10.1093/ije/dyad124

Regulatory circuits of mitophagy restrict distinct modes of cell death during memory CD8 < sup > + < /sup > T cell formation
Sci Immunol. 2023 Sep 29;8(87):eadf7579. doi: 10.1126/sciimmunol.adf7579. Epub 2023 Sep 22.ABSTRACTMitophagy, a central process guarding mitochondrial quality, is commonly impaired in human diseases such as Parkinson's disease, but its impact in adaptive immunity remains unclear. The differentiation and survival of memory CD8+ T cells rely on oxidative metabolism, a process that requires robust mitochondrial quality control. Here, we found that Parkinson's disease patients have a reduced frequency of CD8+ memory T cells compared with healthy donors and failed to form memory T cells upon vaccination against COVID-19, highli...

Taxane-Based Chemotherapy Is Effective in Metastatic Appendiceal Adenocarcinoma
Oncologist. 2023 Sep 20:oyad263. doi: 10.1093/oncolo/oyad263. Online ahead of print.ABSTRACTAppendiceal cancer is a rare, orphan disease with no therapies currently approved by the FDA for its treatment. Given the limited data regarding drug efficacy, these tumors have historically been treated with chemotherapy designed for colon cancer. However, an overwhelming body of molecular data has demonstrated that appendiceal adenocarcinoma is a distinct entity with key molecular differences from colon cancer, notably rare APC mutation. Recognizing that APC loss-of-function is thought to contribute to taxane resistance and that t...

ctDNA-guided adjuvant treatment after radical-intent treatment of metastatic spread from colorectal cancer-the first interim results from the OPTIMISE study
CONCLUSION: The study was proven to be feasible and continues in the planned large-scale phase II trial. Results from the OPTIMISE study will potentially optimize the adjuvant treatment of patients undergoing radical-intent treatment of mCRC, thereby improving survival and reducing chemotherapy-related toxicity.PMID:37738268 | DOI:10.1080/0284186X.2023.2259083

Recommendations on data sharing in HIV drug resistance research
PLoS Med. 2023 Sep 22;20(9):e1004293. doi: 10.1371/journal.pmed.1004293. Online ahead of print.ABSTRACTAuthor summary • Human immunodeficiency virus (HIV) drug resistance has implications for antiretroviral treatment strategies and for containing the HIV pandemic because the development of HIV drug resistance leads to the requirement for antiretroviral drugs that may be less effective, less well-tolerated, and more expensive than those used in first-line regimens. • HIV drug resistance studies are designed to determine which HIV mutations are selected by antiretroviral drugs and, in turn, how these mutations affect ant...

Large-scale whole exome sequencing studies identify two genes,CTSL and APOE, associated with lung cancer
In this study, we investigate the contribution of rare variants (RVs) (minor allele frequency <0.01) to lung cancer through two large whole exome sequencing case-control studies. We first performed gene-based association tests using a novel Bayes Factor statistic in the International Lung Cancer Consortium, the discovery study (European, 1042 cases vs. 881 controls). The top genes identified are further assessed in the UK Biobank (European, 630 cases vs. 172 864 controls), the replication study. After controlling for the false discovery rate, we found two genes, CTSL and APOE, significantly associated with lung cancer i...

Decentralizing cancer care in sub-Saharan Africa through an integrated regional cancer centre model: The case of Kenya
This study aimed to analyze the status of the utilization of services at regional cancer centers. We analyzed data from the district health information system, focusing on patient demographics, visit type, cancer stage, and the type of treatment provided. For comparison, a trend analysis of new cancer cases recorded at the main national referral hospital between 2011-2021 was also conducted. We conducted a descriptive analysis of the variables of interest; the median was used to summarize continuous variables and percentages were used for categorical variables. A total of 29,321 patients visited the regional centers in 202...

Risk of thromboembolic events associated with different multiple myeloma regimens in Taiwan: a nested case-control study
J Thromb Thrombolysis. 2023 Sep 22. doi: 10.1007/s11239-023-02887-7. Online ahead of print.ABSTRACTAlthough novel agents for multiple myeloma (MM) have a better response rate and survival in both newly diagnosed and relapsed/refractory MM patients, concerns regarding the association between MM treatments and thromboembolic events have been raised. The aim of this population-based study was to examine the association between different combinations of MM treatments and the risk of thromboembolic events. We conducted a nested case-control study using the Taiwan Cancer Registry (TCR) and National Health Insurance Research Data...

Peroxisomal ROS control cytosolic Mycobacterium tuberculosis replication in human macrophages
J Cell Biol. 2023 Dec 4;222(12):e202303066. doi: 10.1083/jcb.202303066. Epub 2023 Sep 22.ABSTRACTPeroxisomes are organelles involved in many metabolic processes including lipid metabolism, reactive oxygen species (ROS) turnover, and antimicrobial immune responses. However, the cellular mechanisms by which peroxisomes contribute to bacterial elimination in macrophages remain elusive. Here, we investigated peroxisome function in iPSC-derived human macrophages (iPSDM) during infection with Mycobacterium tuberculosis (Mtb). We discovered that Mtb-triggered peroxisome biogenesis requires the ESX-1 type 7 secretion system, criti...

The impact of intensified staging and combined therapies in locally advanced pancreatic cancer: a secondary analysis of prospective studies
CONCLUSIONS: The use of comprehensive pre-treatment staging, including laparoscopy and 18F-FDG-PET/CT scan, is an effective approach in identifying occult metastatic disease in LAPC patients. Our findings offer valuable insights into accurate staging and treatment efficacy, providing evidence-based support for optimal management strategies in LAPC patients.PMID:37737898 | DOI:10.1097/JS9.0000000000000755
