Peroxisomal ROS control cytosolic Mycobacterium tuberculosis replication in human macrophages
J Cell Biol. 2023 Dec 4;222(12):e202303066. doi: 10.1083/jcb.202303066. Epub 2023 Sep 22.ABSTRACTPeroxisomes are organelles involved in many metabolic processes including lipid metabolism, reactive oxygen species (ROS) turnover, and antimicrobial immune responses. However, the cellular mechanisms by which peroxisomes contribute to bacterial elimination in macrophages remain elusive. Here, we investigated peroxisome function in iPSC-derived human macrophages (iPSDM) during infection with Mycobacterium tuberculosis (Mtb). We discovered that Mtb-triggered peroxisome biogenesis requires the ESX-1 type 7 secretion system, criti...

The impact of intensified staging and combined therapies in locally advanced pancreatic cancer: a secondary analysis of prospective studies
CONCLUSIONS: The use of comprehensive pre-treatment staging, including laparoscopy and 18F-FDG-PET/CT scan, is an effective approach in identifying occult metastatic disease in LAPC patients. Our findings offer valuable insights into accurate staging and treatment efficacy, providing evidence-based support for optimal management strategies in LAPC patients.PMID:37737898 | DOI:10.1097/JS9.0000000000000755

Harnessing Dendritic Cells: Next Frontier for Durable Immune Control in Myeloma
Clin Cancer Res. 2023 Sep 22:OF1-OF3. doi: 10.1158/1078-0432.CCR-23-2292. Online ahead of print.ABSTRACTImmune-based approaches including T-cell redirection have transformed the therapeutic landscape in myeloma. Injection of dendritic cells (DC) led to the induction of immune responses in vaccinated patients with myeloma. These studies pave the way for future combination strategies harnessing DCs to enhance tumor immunity and improve outcomes in myeloma. Prevalence and predictors of colorectal cancer screening in the United States: evidence from the HINTS database 2018 to 2020
CONCLUSION: CRC screening rates among adults 45-49 years are low but are increasing steadily. Odds of CRC screening among Blacks is high which is encouraging while the odds among current smokers is low and concerning given their increased risk of developing CRC.PMID:37737304 | DOI:10.1007/s10552-023-01795-8

Association between weight loss agents and elevated liver enzymes: a population-based cross-sectional study
This study aimed to investigate the associations between weight loss agents and elevated liver enzymes at the population-level. We conducted a cross-sectional study using Korea National Health and Nutrition Examination Survey (KNHANES) data from 2013 to 2019. This study included 36,259 participants over 20 years of age who completed the questionnaire and had no history of hepatitis, cancer, or renal failure. In these participants, we analyzed associations between weight loss agents and elevated liver enzymes by constructing multiple logistic regression models with adjustment for confounding factors and stratified by sex, a...

Fetal Exposure to Preeclampsia and Later Risk of Cardiometabolic Disorders: A Population-Based Cohort Study
CONCLUSIONS: Fetal preeclampsia exposure was associated with adult life hypertension, diabetes, and dyslipidemia, but these associations were likely due to shared etiological factors, rather than exposure to the preeclamptic condition itself.PMID:37737002 | DOI:10.1161/HYPERTENSIONAHA.122.20682

Oncolytic virus oHSV2 combined with PD-1/PD-L1 inhibitors exert antitumor activity by mediating CD4  + T and CD8 + T cell infiltration in the lymphoma tumor microenvironment
Autoimmunity. 2023 Dec;56(1):2259126. doi: 10.1080/08916934.2023.2259126. Epub 2023 Sep 22.ABSTRACTA novel therapeutic regimen showed that the oncolytic type II herpes simplex virus (oHSV2) was able to prevent colorectal cancer growth, recurrence, and metastasis. However, no study has yet explored whether oHSV2 has an impact on the development of diffuse large B-cell lymphoma (DLBCL). We chose the clinical chemotherapeutic drug doxorubicin (DOX) as a positive control to evaluate the effect of oHSV2 infection on the apoptotic, invasive, and proliferative capacity of DLBCL cells. We next further explored the therapeutic effi...

Argon-helium knife cryoablation plus programmed cell death protein 1 inhibitor in the treatment of advanced soft tissue sarcomas: there is no evidence of the synergistic effects of this combination therapy
CONCLUSION: Cryoablation combined with PD-1 inhibitors in the therapy of advanced STS is safe and can effectively shrink the cryoablation-target lesion. However, there is no evidence of the synergistic effects of this combination therapy.PMID:37736543 | PMC:PMC10509548 | DOI:10.3389/fonc.2023.1185291

Automated radiosynthesis of < sup > 18 < /sup > FFMPEP- < em > d < /em > < sub > 2 < /sub > for cannabinoid receptor PET imaging
Am J Nucl Med Mol Imaging. 2023 Aug 15;13(4):171-178. eCollection 2023.ABSTRACTThe cannabinoid subtype 1 receptor (CB1R) is highly expressed in the central nervous system and abnormalities in regional CB1R density are associated with neurodegenerative disorders. The PET tracer [18F]FMPEP-d2 is an inverse CB1R agonist which was shown to be suitable for non-invasive PET imaging. In this work, we reported the fully automated radiosynthesis of [18F]FMPEP-d2 on a Synthra RNplus research module. In a total synthesis time of 70 min, [18F]FMPEP-d2 was obtained in 2.2 ± 0.1 GBq (n = 3) with excellent radiochemical and chemical pur...
