Gene Correction of Human Induced Pluripotent Stem Cells Repairs the Cellular Phenotype in Pulmonary Alveolar Proteinosis.
Conclusions: These data establish PAP-iPSC-derived monocytes/macrophages as a valid in vitro disease model of CSF2RA-deficient PAP, and introduce gene-corrected iPSC-derived monocytes/macrophages as a potential autologous cell source for innovative therapeutic strategies. Macrolide Antibiotics and the Risk of Cardiac Arrhythmias.
Abstract Randomized, controlled trials have demonstrated that chronic therapy with macrolide antibiotics reduces the morbidity of patients with cystic fibrosis, non-cystic fibrosis bronchiectasis, chronic obstructive pulmonary disease and nontuberculous mycobacterial infections. Lower levels of evidence indicate that chronic macrolides are also effective in patients with panbronchiolitis, and bronchiolitis obliterans and rejection after lung transplant. Genotype-Phenotype Correlations for Infants and Children with ABCA3 Deficiency.
Conclusions: Genotype-phenotype correlations exist for homozygous or compound heterozygous mutations in ABCA3. Frameshift or nonsense ABCA3 mutations are predictive of neonatal presentation and poor outcome, while missense, splice site, and insertion/deletions are less reliably associated with age of presentation and prognosis. Wnt Co-receptor Lrp5 is a Driver of Idiopathic Pulmonary Fibrosis.
Conclusions: We show that the Wnt co-receptor, Lrp5, is a genetic driver of lung fibrosis in mice and a marker of disease progression and severity in humans with IPF. Evidence that TGFβ signaling can override a loss in Lrp5 has implications for patient selection and timing of Wnt pathway inhibitors in lung fibrosis.
