Role of serum ‐ and glucocorticoid‐inducible kinases in stroke
Abstract Increased expression of serum‐ and glucocorticoid‐inducible kinase 1 (SGK1) can be induced by stress and growth factors in mammals, and plays an important role in cancer, diabetes, and hypertension. A recent work suggested that SGK1 activity restores damage in a stroke model. To further investigate the role of SGKs in ischemic brain injury, we examined how SGK inhibitors influence stroke outcome in vivo and neurotoxicity in vitro. Infarct volumes were compared in adult mice with middle cerebral artery occlusion, followed by 24 h reperfusion, in the absence or presence of SGK inhibitors. Neurotoxicity assay,...

MMP‐9 in translation: from molecule to brain physiology, pathology, and therapy
This article is part of the 60th Anniversary special issue. MMP‐9, through cleavage of specific target proteins, plays a major role in synaptic plasticity and neuroinflammation, and by those virtues contributes to brain physiology and a host of neurological and psychiatric disorders. This article is part of the 60th Anniversary special issue.

Attenuation of cerebral ischemic injury in interferon regulatory factor 3‐deficient rat
In this study, a novel IRF3 knockout rat strain and a transgenic rat strain with neuronal‐specific IRF3 over‐expression (IRF3‐TG) were created. Subsequently, the generated IRF3‐knockout rats, the neuronal‐specific IRF3 over‐expressing rats and their corresponding controls were subjected to transient middle cerebral artery occlusion and followed by reperfusion, to investigate the exact role of IRF3 in cerebral I/R in rats. In contrast to the results in mice, IRF3 deficiency in rats provided significant protection against cerebral I/R injury and inhibited neuronal apoptosis, inflammation, and oxidative stress aft...

Plasmatic retinol‐binding protein 4 and glial fibrillary acidic protein as biomarkers to differentiate ischemic stroke and intracerebral hemorrhage
We describe the discovery and subsequent replications of RBP4 and its combination with circulating GFAP as plasmatic biomarkers for hyperacute stroke subtype differentiation. The combination of these biomarkers and others might aid to speed up the discrimination of both stroke subtypes improving the outcome of patients. A rapid differentiation of ischemic stroke from intracerebral hemorrhage is essential to provide the appropriate treatment. We describe the discovery and subsequent replications of RBP4 and its combination with circulating GFAP as plasmatic biomarkers for hyperacute stroke subtype differentiation. The comb...

IL‐1alpha induces angiogenesis in brain endothelial cells in vitro: implications for brain angiogenesis after acute injury
In conclusion, our data highlights a potential new role for IL‐1 in brain repair mechanisms and identifies IL‐1α as a potential new therapy to promote post‐stroke angiogenesis. Inflammation is a major contributor to neuronal injury and is associated with poor outcome after neurotrauma. We demonstrate that cytokine IL‐1α is chronically elevated in the brain after experimental stroke suggesting that it is present chronically post‐stroke. We demonstrate that IL‐1α is a potent mediator of endothelial activation and inducer of angiogenic markers in endothelial cells. Our data highlights a new role for IL‐1 in ...
