A CRISPR-Cas12a-based platform facilitates the detection and serotyping of Streptococcus suis serotype 2
In this study, we developed a high-fidelity detection and serotyping platform for S. suis serotype 2 based on recombinase polymerase amplification (RPA) and a clustered regularly interspaced short palindromic repeat (CRISPR)-Cas12a system called Cards-SSJ/K. Cards-SSJ had a detection limit of 10 CFU, takes <60 min, and no cross-reaction was found with other S. suis serotypes, closely related Streptococcus spp., or common pig pathogens, and Cards-SSK could differentiate serotype 2 from serotype 1/2. Results from Cards-SSJ and qPCR were equivalent in detecting S. suis serotype 2 in tissue samples. Analysis indicated that ...

Amorphous amEu-NH < sub > 2 < /sub > BDC and amTb-NH < sub > 2 < /sub > BDC as ratio fluorescence probes for smartphone-integrated naked eye detection of bacillus anthracis biomarker
Talanta. 2023 Sep 14;267:125164. doi: 10.1016/j.talanta.2023.125164. Online ahead of print.ABSTRACTThe abnormal concentration of anthrax spore biomarker 2,6-pyridinedicarboxylic acid (2,6-DPA) will seriously affect public health. Therefore, a sensitive and rapid assay for 2,6-DPA monitoring is of vital importance. In this work, novel nano-sized amorphous Eu-NH2BDC (amEu-NH2BDC) and amorphous Tb-NH2BDC (amTb-NH2BDC) metal organic frameworks are prepared by adjusting the ratio of metal and ligand, respectively. Both of them exhibit highly sensitive and selective ratiometric fluorescence detection for 2,6-DPA with wider linea...

The performance of hypochlorous acid modified Ag nanoparticle-based assay in the determination of total antioxidant capacity of Boswellia Serrata and Aronia
In this study, the SNPAC method was developed by using hypochlorous (ClO-) at pH 9.3. Polyvinylpyrrolidone (PVP) was used as a stabilizer. The existence of HOCl enables to formation stronger and more stable yellow emission which gives sensitivity. As a result, lower LOD and LOQ values were achieved by the AgNP-HOCl method. The obtaining lower values was attributed to the reoxidation of silver atoms with ClO- ions in the presence of PVP. The newly modified method was applied to the total antioxidant capacity (TAC) measurement of Boswellia Serrata, Aronia Melanocarpa tea, and juice. The results were compared with the results...

Liquid chromatographic retention time prediction models to secure and improve the feature annotation process in high-resolution mass spectrometry
Talanta. 2023 Sep 17;267:125214. doi: 10.1016/j.talanta.2023.125214. Online ahead of print.ABSTRACTThe development of quantitative structure-retention relationship (QSRR) models has, until recently, required an adequate selection of molecular descriptors necessarily obtained based on a known chemical structure. However, these complex descriptors are not always available nor calculable when the high-resolution mass spectrometry (HRMS) annotation process is underway. Depending on the level of annotation, many structures or even various molecular formulas could be candidates. To secure and improve the annotation process and t...

A plasmonic gold nano-surface functionalized with the estrogen receptor for fast and highly sensitive detection of nanoplastics
Talanta. 2023 Sep 16;267:125211. doi: 10.1016/j.talanta.2023.125211. Online ahead of print.ABSTRACTNanoplastics are a global emerging environmental problem whose effects might pose potential threats to the human's health. Despite the relevance of the issue, fast, reliable and quantitative in situ analytical approaches to determine nanoplastics are not yet available. The aim of this work was to devise an optical sensor with the goal of direct detecting and quantifying nanoplastics in seawater without sample pre-treatments. To this purpose, a nano-plasmonic biosensor was developed by exploiting an Estrogen Receptor (ER) reco...

Polyazine nanoparticles as anchors of PQQ glucose dehydrogenase for its most efficient bioelectrocatalysis
We report on the drop-cast production of glucose biosensors based on the most efficient bioelectrocatalysis by pyrroloquinoline quinone dependent glucose dehydrogenase (PQQ GDH). To orient the enzyme upon immobilization we suggest using poly(Methylene Green) (p(MG)) nanoparticles acting as anchors. Synthesis of polymeric anchors has been carried out in course of Methylene Green electropolymerization, which allowed to tune polymer-to-monomer ratio in the drop-cast mixtures varying the monomer concentration and applying different number of potential sweep cycles. Except for elimination of electrochemical step for the electro...

A luminescent MOF-based nonenzymatic probe for colorimetric/photothermal/fluorescence triple-mode assay of uric acid in body fluids
Talanta. 2023 Sep 15;267:125201. doi: 10.1016/j.talanta.2023.125201. Online ahead of print.ABSTRACTMonitoring the levels of uric acid (UA) in body fluids is of great significance in the clinical diagnosis and therapy of related diseases. Herein, a novel nanocomposite R6G@Fe-MOF based nonenzymatic probe is presented to provide a ratiometric fluorescent, colorimetric, and photothermal triple read-out signal for the visual, sensitive, and convenient assay of UA. The framework structure of the in situ encapsulated R6G@Fe-MOF is found to decompose upon the addition of UA, resulting in the reduction of Fe3+ to Fe2+. This reducti...

Engineering an activatable fluorescent probe for studying ONOO < sup > - < /sup > in pyroptotic process
Talanta. 2023 Sep 16;267:125216. doi: 10.1016/j.talanta.2023.125216. Online ahead of print.ABSTRACTPyroptosis, a recently discovered form of programmed cell death, plays a pivotal role in oncological treatment. Howbeit, the mechanisms underlying pyroptosis in tumor treatment remain unclear. Previous research has demonstrated that the occurrence of pyroptosis generally accompanies a surge of reactive oxygen species (ROS) generation, with ONOO- being one of these ROS and closely linked to numerous diseases. Therefore, it is imperative to investigate the potential association between ONOO- and pyroptosis. Herein, a highly sen...

SERS-based methods for the detection of genomic biomarkers of cancer
Talanta. 2023 Sep 15;267:125198. doi: 10.1016/j.talanta.2023.125198. Online ahead of print.ABSTRACTGenomic biomarkers of cancer are based on changes in nucleic acids, which include abnormal expression levels of some miRNAs, point mutations in DNA sequences, and altered levels of DNA methylation. The presence of tumor-related nucleic acids in body fluids (blood, saliva, or urine) makes it possible to achieve a non-invasive early-stage cancer diagnosis. Currently existing techniques for the discovery of nucleic acids require complex, time-consuming, costly assays and have limited multiplexing abilities. Surface-enhanced Rama...
