Skeletal muscle function is altered in male mice on low dose androgen receptor antagonist or estrogen receptor agonist
We examined the effect of acute (2 week) exposures to 17α-ethinyl estradiol (EE2), an estrogen receptor (ER) agonist, or flutamide, an androgen receptor (AR) antagonist, on the contractile function of individual skeletal muscle fibers from slow-contracting soleus and fast-contracting extensor digitorum longus muscles from adult male mice. Single fiber specific tension (force divided by cross-sectional area) was decreased with flutamide treatment in all myosin heavy chain fiber types examined (MHC I, IIA and IIB); similar effects were observed with EE2-treatment but only in the fastest-contracting MHC IIB fibers. The decre...

"Longitudinal course of sex steroids from pregnancy to postpartum"
CONCLUSIONS: A comprehensive understanding of the influence of SS levels on associated maternal and fetal pathologies is currently hindered by two main factors. First, reported SS levels vary widely during the peripartum period. Second, the current state of knowledge on how SS are associated with pathologies in mothers and babies is largely based on correlational studies, and causality thus remains unclear. Consequently, we recommend the development of a systematic reference framework that follows the suggestions presented in this review. This would enable the establishment of SS reference values for a healthy population, ...

Leptin, but not Estradiol, Signaling in PACAP Neurons Modulates Puberty Onset
Endocrinology. 2023 Jun 26;164(8):bqad097. doi: 10.1210/endocr/bqad097.ABSTRACTThe adipose-derived hormone leptin critically modulates reproductive function, such that its absence results in hypothalamic hypogonadism. Pituitary adenylate cyclase-activating polypeptide (PACAP)-expressing neurons are potential mediators of leptin's action on the neuroendocrine reproductive axis because they are leptin-sensitive and involved in both feeding behavior and reproductive function. In the complete absence of PACAP, male and female mice exhibit metabolic and reproductive abnormalities, yet there is some sexual dimorphism in the repr...

RFamide-related peptide 3 signaling via neuropeptide FF receptor stimulates prolactin secretion in female rats
Endocrinology. 2023 Jul 4:bqad102. doi: 10.1210/endocr/bqad102. Online ahead of print.ABSTRACTThe RF-amide peptides comprise a family of neuropeptides that includes the kisspeptin (Kp), the natural ligand of kisspeptin receptor (Kiss1r), and the RFamide-related peptide 3 (RFRP-3) that binds preferentially to the neuropeptide FF receptor 1 (Npffr1). Kp stimulates prolactin (PRL) secretion through the inhibition of tuberoinfundibular dopaminergic (TIDA) neurons. Because Kp has also affinity to Npffr1, we investigated the role of Npffr1 in the control of PRL secretion by Kp and RFRP-3. Intracerebroventricular (i.c.v.) injecti...

Organophosphate Esters Disrupt Steroidogenesis in KGN Human Ovarian Granulosa Cells
Endocrinology. 2023 Jun 8:bqad089. doi: 10.1210/endocr/bqad089. Online ahead of print.ABSTRACTOrganophosphate esters (OPEs) are used extensively as flame retardants and plasticizers and are found ubiquitously in the environment and human matrices. Previous studies suggested that exposure to some of these chemicals may disrupt the homeostasis of female sex hormones and have detrimental effects on female fertility. Here, we determined the effects of OPEs on the function of KGN ovarian granulosa cells. We hypothesized that OPEs alter the steroidogenic ability of these cells by dysregulating the expression of transcripts invol...
