DKK1 Affects Survival of Patients with Head and Neck Squamous Cell Carcinoma by Inducing Resistance to Radiotherapy and Immunotherapy
CONCLUSION: Our data show that DKK1 can affect both radiotherapy and immunotherapy in HNSC, suggesting that DKK1 can be a potential target for radioimmunology in HNSC.PMID:36690301 | DOI:10.1016/j.radonc.2023.109485

Targeting HER3-dependent activation of nuclear AKT improves radiotherapy of non-small celllung cancer
CONCLUSION: IR-induced activation of nuclear AKT occurs inside the nucleus that is mainly dependent on HER3 expression in NSCLC. These findings suggest that targeting HER3 in combination with radiotherapy may provide a logical treatment option for investigation in selected NSCLC patients.PMID:35839938 | DOI:10.1016/j.radonc.2022.07.008

Cancer cell motility is affected through 3D cell culturing and SCF/c-Kit pathway but not by X-irradiation.
CONCLUSION: X-irradiation affects neither the expression of important EMT genes such as vimentin, E-cadherin and N-cadherin nor SCF/c-Kit signaling and, as a consequence, does not alter cell motility. PMID: 27178146 [PubMed - as supplied by publisher]

Quantitative proteomics unveiled: Regulation of DNA double strand break repair by EGFR involves PARP1.
CONCLUSION: We have established a powerful, quantitative phosphoproteomic approach to investigate regulatory mechanisms in DSB repair, dependent on protein phosphorylation after irradiation. Using this approach we have identified PARP1 as a mediator of EGFR/MEK-dependent regulation of DSB repair. PMID: 26422459 [PubMed - as supplied by publisher]

Nuclear EGFR renders cells radio-resistant by binding mRNA species and triggering a metabolic switch to increase lactate production.
CONCLUSIONS: We showed that nuclear EGFR selectively binds and stabilizes mRNA involved in the Warburg effect in response to irradiation. As a consequence, cells switch from aerobic to anaerobic glucose metabolism, which can be prevented by HIF1α inhibitor BAY87-2243, Dasatinib, Erlotinib or EGFR siRNA. AMPK-independent autophagy promotes radioresistance of human tumor cells under clinical relevant hypoxia in vitro.
CONCLUSION: Under clinically relevant hypoxia (1% O2) the stimulation of autophagy mediates resistance of hypoxic tumor cells to ionizing radiation, which is independent of AMPK signaling. PMID: 26318663 [PubMed - as supplied by publisher]

Radiosensitization of NSCLC cells by EGFR inhibition is the result of an enhanced p53-dependent G1 arrest.
CONCLUSION: For NSCLC cells increase in radiosensitivity by EGFR inhibition results from enhanced G1-arrest. However, this effect does not lead to improved tumor control because cells can be released from this arrest by re-stimulation. PMID: 25796091 [PubMed - as supplied by publisher]

The PERK/ATF4/LAMP3-arm of the unfolded protein response affects radioresistance by interfering with the DNA damage response.
CONCLUSIONS: The PERK/ATF4/LAMP3-arm causes radioresistance of breast cancer cells by increasing DNA damage repair signalling. Inhibition of PERK and/or autophagy may sensitise tumours to radiotherapy. PMID: 23891100 [PubMed - as supplied by publisher]

Osteopontin and splice variant expression level in human malignant glioma: Radiobiologic effects and prognosis after radiotherapy.
CONCLUSIONS: OPNb is partially able to compensate the effects of OPNa and OPNc knockdown in U251MG cells. High OPN plasma levels at the end of radiotherapy are associated with poor survival. PMID: 23891093 [PubMed - as supplied by publisher]

Acquired resistance to cetuximab is associated with the overexpression of Ras family members and the loss of radiosensitization in head and neck cancer cells.
CONCLUSION: Targeting Ras and PI3K activity improves the outcome of irradiation in cetuximab-resistant HNSCC cell lines in vitro. PMID: 23891090 [PubMed - as supplied by publisher]
