Irisin protects against sepsis-associated encephalopathy by suppressing ferroptosis via activation of the Nrf2/GPX4 signal axis
In conclusion, irisin could ameliorate inflammatory microenvironment in SAE by suppressing hippocampus ferroptosis via the Nrf2/GPX4 signaling pathway.PMID:35660523 | DOI:10.1016/j.freeradbiomed.2022.05.023

Hydrogen sulfide alleviates particulate matter-induced emphysema and airway inflammation by suppressing ferroptosis
CONCLUSION: This research suggested that H2S alleviated PM-induced emphysema and airway inflammation via restoring redox balance and inhibiting ferroptosis through regulating Nrf2-PPAR-ferritinophagy signaling pathway.PMID:35490984 | DOI:10.1016/j.freeradbiomed.2022.04.014

Old targets, new strategy: Apigenin-7-O- β-d-(-6″-p-coumaroyl)-glucopyranoside prevents endothelial ferroptosis and alleviates intestinal ischemia-reperfusion injury through HO-1 and MAO-B inhibition
This study aimed to investigate the effects of APG on IIRI both in vivo and in vitro and identify the potential molecular mechanism. We found that APG could significantly improve intestinal edema and increase Chiu's score. MST analysis suggested that APG could specifically bind to heme oxygenase 1 (HO-1) and monoamine oxidase b (MAO-B). Simultaneously, APG could attenuate ROS generation and Fe2+ accumulation, maintain mitochondria function thus inhibit ferroptosis with a dose-dependent manner. Moreover, we used siRNA silencing technology to confirm that knocking down both HO-1 and MAO-B had a positive effect on intestine. ...

Role of selenoprotein P expression in the function of pancreatic β cells: Prevention of ferroptosis-like cell death and stress-induced nascent granule degradation
Free Radic Biol Med. 2022 Mar 19:S0891-5849(22)00104-6. doi: 10.1016/j.freeradbiomed.2022.03.009. Online ahead of print.ABSTRACTSelenoprotein P (SELENOP) is a major selenium (Se)-containing protein (selenoprotein) in human plasma that is mainly synthesized in the liver. SELENOP transports Se to the cells, while SELENOP synthesized in peripheral tissues is incorporated in a paracrine/autocrine manner to maintain the levels of cellular selenoproteins, called the SELENOP cycle. Pancreatic β cells, responsible for the synthesis and secretion of insulin, are known to express SELENOP. Here, using MIN6 cells as a mouse model for...

Elabela alleviates ferroptosis, myocardial remodeling, fibrosis and heart dysfunction in hypertensive mice by modulating the IL-6/STAT3/GPX4 signaling
In conclusion, elabela antagonizes Ang II-mediated promotion of CMVECs ferroptosis, adverse myocardial remodeling, fibrosis and heart dysfunction through modulating the IL-6/STAT3/GPX4 signaling pathway. Targeting elabela-APJ axis serves as a novel strategy for hypertensive heart diseases.PMID:35122997 | DOI:10.1016/j.freeradbiomed.2022.01.020

Novel mechanistic insight on the neuroprotective effect of berberine: The role of PPAR δ for antioxidant action
Free Radic Biol Med. 2022 Jan 27:S0891-5849(22)00033-8. doi: 10.1016/j.freeradbiomed.2022.01.022. Online ahead of print.ABSTRACTCerebral ischemic stroke ranks the second leading cause of death and the third leading cause of disability in lifetime all around the world, urgently necessitating effective therapeutic interventions. Reactive oxygen species (ROS) have been implicated in stroke pathogenesis and peroxisome proliferator-activated receptors (PPARs) are prominent targets for ROS management. Although recent research has shown antioxidant effect of berberine (BBR), little is known regarding its effect upon ROS-PPARs sig...

SESN2/NRF2 signaling activates as a direct downstream regulator of the PERK pathway against endoplasmic reticulum stress to improve the in vitro maturation of porcine oocytes
Free Radic Biol Med. 2021 Dec 16:S0891-5849(21)01110-2. doi: 10.1016/j.freeradbiomed.2021.12.258. Online ahead of print.ABSTRACTNuclear erythroid 2-related factor 2 (NRF2) is a critical regulator of oxidative stress in mammalian oocytes. Our previous study described the protective effects of Sestrin-2 (SESN2) as a stress regulator against endoplasmic reticulum (ER) stress in porcine oocytes during in vitro maturation (IVM). However, their roles in unfolded protein response-related signaling pathways in porcine oocyte maturation capacity remain unknown. The purpose of this study was to evaluate the role of SESN2/NRF2 signal...
