Quantitative Fluorescence Spectral Analysis of Protein Denaturation

This chapter describes a procedure of global analysis of the steady-state spectra measured with different concentrations of the denaturant to quantitatively study protein denaturation. With the help of physicochemical models, relevant spectral parameters that characterize the folding intermediate and thermodynamic parameters that describe a three-state model N  $$ \Leftrightarrow $$  I  $$ \Leftrightarrow $$  U can be estimated.
