Palbociclib releases the latent differentiation capacity of neuroblastoma cells
Dev Cell. 2023 Sep 9:S1534-5807(23)00443-4. doi: 10.1016/j.devcel.2023.08.028. Online ahead of print.ABSTRACTNeuroblastoma is the most common extracranial solid tumor in infants, arising from developmentally stalled neural crest-derived cells. Driving tumor differentiation is a promising therapeutic approach for this devastating disease. Here, we show that the CDK4/6 inhibitor palbociclib not only inhibits proliferation but induces extensive neuronal differentiation of adrenergic neuroblastoma cells. Palbociclib-mediated differentiation is manifested by extensive phenotypic and transcriptional changes accompanied by the es...

Apical polarity and actomyosin dynamics control Kibra subcellular localization and function in Drosophila Hippo signaling
This study provides a mechanistic understanding of the relationship between the Hippo pathway, polarity, and actomyosin cytoskeleton, and it offers novel insights into how fundamental features of epithelial tissue architecture can serve as inputs into signaling cascades that control tissue growth, patterning, and morphogenesis.PMID:37729921 | DOI:10.1016/j.devcel.2023.08.029

Phagocytosis by macrophages promotes pancreatic β cell mass reduction after parturition in mice
Dev Cell. 2023 Aug 19:S1534-5807(23)00398-2. doi: 10.1016/j.devcel.2023.08.002. Online ahead of print.ABSTRACTElucidating the mechanism(s) modulating appropriate tissue size is a critical biological issue. Pancreatic β cells increase during pregnancy via cellular proliferation, but how β cells promptly decrease to the original amount after parturition remains unclear. Herein, we demonstrate the role and mechanism of macrophage accumulation in this process. In the final stage of pregnancy, HTR1D signaling upregulates murine β cell CXCL10, thereby promoting macrophage accumulation in pancreatic islets via the CXCL10-CXCR3...

Chromatin remodeling of prostaglandin signaling in smooth muscle enables mouse embryo passage through the female reproductive tract
Dev Cell. 2023 Sep 7:S1534-5807(23)00440-9. doi: 10.1016/j.devcel.2023.08.025. Online ahead of print.ABSTRACTEarly mammalian development occurs during embryo transit of the female reproductive tract. Transport is orchestrated by secreted oviduct fluid, unidirectional beating of epithelial cilia, and smooth muscle contractions. Using gene-edited mice, we document that conditional disruption of a component of the SWI/SNF chromatin remodeling complex in smooth muscle cells prevents transport through the oviduct without perturbing embryogenesis. Analysis with RNA sequencing (RNA-seq), transposase-accessible chromatin with sequ...

An improved Erk biosensor detects oscillatory Erk dynamics driven by mitotic erasure during early development
Dev Cell. 2023 Sep 9:S1534-5807(23)00436-7. doi: 10.1016/j.devcel.2023.08.021. Online ahead of print.ABSTRACTExtracellular signal-regulated kinase (Erk) signaling dynamics elicit distinct cellular responses in a variety of contexts. The early zebrafish embryo is an ideal model to explore the role of Erk signaling dynamics in vivo, as a gradient of activated diphosphorylated Erk (P-Erk) is induced by fibroblast growth factor (Fgf) signaling at the blastula margin. Here, we describe an improved Erk-specific biosensor, which we term modified Erk kinase translocation reporter (modErk-KTR). We demonstrate the utility of this bi...

Mitochondrial GTP metabolism controls reproductive aging in C.  elegans
We report that mitochondrial GTP (mtGTP) metabolism acts through mitochondrial dynamics factors to regulate reproductive aging. We discovered that germline-only inactivation of GTP- but not ATP-specific succinyl-CoA synthetase (SCS) promotes reproductive longevity in Caenorhabditis elegans. We further identified an age-associated increase in mitochondrial clustering surrounding oocyte nuclei, which is attenuated by GTP-specific SCS inactivation. Germline-only induction of mitochondrial fission factors sufficiently promotes mitochondrial dispersion and reproductive longevity. Moreover, we discovered that bacterial inputs af...

Centrins control chicken cone cell lipid droplet dynamics through lipid-droplet-localized SPDL1
Dev Cell. 2023 Sep 2:S1534-5807(23)00408-2. doi: 10.1016/j.devcel.2023.08.012. Online ahead of print.ABSTRACTAs evolutionarily conserved organelles, lipid droplets (LDs) carry out numerous functions and have various subcellular localizations in different cell types and species. In avian cone cells, there is a single apically localized LD. We demonstrated that CIDEA (cell death inducing DFFA like effector a) and microtubules promote the formation of the single LD in chicken cone cells. Centrins, which are well-known centriole proteins, target to the cone cell LD via their C-terminal calcium-binding domains. Centrins localiz...

Peripheral but critical: Translation of germ granule mRNAs in zebrafish
Dev Cell. 2023 Sep 11;58(17):1517-1518. doi: 10.1016/j.devcel.2023.08.008.ABSTRACTRNA granule components occupy distinct positions within granules. However, the significance behind this organization is unclear. In this issue of Developmental Cell, Westerich et al. show that the periphery of the zebrafish germ granules promotes mRNA translation while its interior represses it, which is critical for germ cell establishment.PMID:37699335 | DOI:10.1016/j.devcel.2023.08.008

KRAS < sup > G12D < /sup > inhibition in pancreatic cancer: Fas expression facilitates immune clearance
Dev Cell. 2023 Sep 11;58(17):1515-1516. doi: 10.1016/j.devcel.2023.08.020.ABSTRACTIn an article in this issue of Developmental Cell and in a second paper in Cancer Cell, Mahadevan et al. demonstrate that KrasG12D suppression remodels the immunosuppressive microenvironment of KrasG12D pancreatic cancers, recruits activated CD8+ cytotoxic T cells, and epigenetically upregulates Fas expression in cancer cells, leading to tumor clearance via Fas/FasL-mediated apoptosis.PMID:37699334 | DOI:10.1016/j.devcel.2023.08.020
