Circulating miRNA ‐150‐5p is associated with immune‐mediated early β‐cell loss in a humanized mouse model
ConclusionsOur data demonstrated that immune ‐related miRNAs are associated with human islet xenograft rejection in mice. miR‐150‐5p is increased in human islet graft and in the circulation during islet xenograft rejection and β‐cell destruction prior to hyperglycemia and may be an early biomarker for islet xenograft rejection. Chronic dialysis in patients with end ‐stage renal disease: Relevance to kidney xenotransplantation
AbstractRenal allotransplantation clearly offers better survival and quality of life for end ‐stage renal disease (ESRD) patients than chronic dialysis. The median waiting time for a deceased donor kidney in a suitable ESRD patient is 3.9 years. The initial candidates for pig kidney xenotransplantation will be those with ESRD unlikely to receive an allograft within a reasonable period of time. It is thus reasonable to ascertain whether clinical trials of xenotransplantation might likewise offer superior outcomes. Chronic dialysis in patients with ESRD is associated with poor quality of life, significant morbidity, and r...

Amino acid and lipid profiles following pig ‐to‐primate liver xenotransplantation
This report aims to further investigate the post‐LXT lipid and amino acid metabolism profile in our longest surviving recipients (25 and 29 da ys). Experimental samples and control samples, consisting of pre‐transplant porcine and baboon serum and plasma, were analyzed for standard lipid profiles and for amino acid levels. Lipid profiles of LXT recipients remained stable following xenotransplantation compared to donor porcine baseline le vels. Amino acid concentrations also remained similar to baseline controls, with the exception of a 3‐foldincrease inl‐ornithine and more than a 10‐folddecrease inl‐arginine p...

Comparative evaluation of decellularized porcine liver matrices crosslinked with different chemical and natural crosslinking agents
The objective of this study was to compare the scaffold properties of porcine liver ECM crosslinked with different agents (glutaraldehyde, genipin, and quercetin) to find the best strategy for producing a decellularized matrix with optimal and stable characteristics for transplantation and regeneration. The properties examined include mechanical properties, material stability, immunogenicity, and angiogenic capacity. Scaffolds were implanted into the greater omentum of rats, and their abilities to induce immune cell subpopulation invasion and neovascularization were evaluated. The results show that genipin crosslinking of ...

Will donor ‐derived neoplasia be problematic after clinical pig organ or cell xenotransplantation?
AbstractThere is an increased incidence of certain tumors and other neoplastic disease in organ allotransplant recipients receiving immunosuppressive therapy. Following clinical pig organ xenotransplantation, will there be a risk of the development of neoplasia in the pig graft or in other tissues transplanted with it, eg, lymph nodes? The incidence of neoplasia in young slaughterhouse pigs is very low (<0.005%), but in older pigs is largely unknown (as most pigs are killed within the first six months of life). However, lymphosarcoma, nephroblastoma, and melanoma have been reported in pigs. These tumors should be readil...
