Toll-like receptor expression and functional behavior in platelets from patients with systemic lupus erythematosus
CONCLUSION: Our study demonstrates normal platelet TLR expression and function together with increased circulating platelet-monocyte aggregates. In addition, a direct correlation was observed between plasma HMGB1 levels and platelet vWf release following TLR2 stimulation. This platelet behavior in a group of patients undergoing HCQ treatment suggests that platelets could play a role in the inflammatory state of SLE.PMID:38159527 | DOI:10.1016/j.imbio.2023.152782 (Source: Immunobiology)

Complement C1q and von Willebrand factor interaction in atherosclerosis of human carotid artery
Front Immunol. 2023 Dec 14;14:1265387. doi: 10.3389/fimmu.2023.1265387. eCollection 2023.ABSTRACTAtherosclerosis is an inflammatory disease of the vessel wall, with cholesterol crystal (CC) deposition being a hallmark of the disease. As evidence for a cross-talk between complement activation and hemostasis on CC surfaces has been limited to in vitro data, the aim of this study was to demonstrate the presence of C1q-vWF complexes in human atherosclerosis ex vivo. We used immunofluorescence staining and a proximity ligation assay (PLA, Duolink®) to examine the presence, localization, and co-localization of C1q and vWF in fr...

ADAMTS13 inhibits H < sub > 2 < /sub > O < sub > 2 < /sub > -induced human venous endothelial cell injury to attenuate deep-vein thrombosis by blocking the p38/ERK signaling pathway
This study was aimed to explore the role of ADAMTS13 in endothelial cell injury during DVT and the possible mechanism. First, human umbilical vein endothelial cells (HUVECs) were exposed to hydrogen peroxide (H2O2). Then, the mRNA and protein expressions of ADAMTS13 were evaluated with the reverse transcription-quantitative polymerase chain reaction and western blot. After treatment with recombinant ADAMTS13 (rADAMTS13; rA13), the viability and apoptosis of H2O2-induced HUVECs were assessed by cell counting kit-8 assay and terminal-deoxynucleoitidyl transferase-mediated nick end labeling staining. In addition, the levels o...

Reviewing the dysregulation of ADAMTS13 and VWF in sepsis
Shock. 2023 Dec 12. doi: 10.1097/SHK.0000000000002291. Online ahead of print.ABSTRACTSepsis is defined as a life-threatening organ dysfunction caused by excessive host response to infection, and represents the most common cause of in-hospital deaths. Sepsis accounts for 30% of all critically ill patients in the Intensive Care Unit (ICU), and has a global mortality rate of 20%. Activation of blood coagulation during sepsis and septic shock can lead to disseminated intravascular coagulation (DIC) which is characterized by microvascular thrombosis. Von Willebrand Factor and ADAMTS13 are two important regulators of blood coagu...
