Privileged small molecules against neglected tropical diseases: A perspective from structure activity relationships
Eur J Med Chem. 2024 Apr 16;271:116396. doi: 10.1016/j.ejmech.2024.116396. Online ahead of print.ABSTRACTNeglected tropical diseases (NTDs) comprise diverse infections with more incidence in tropical/sub-tropical areas. In spite of preventive and therapeutic achievements, NTDs are yet serious threats to the public health. Epidemiological reports of world health organization (WHO) indicate that more than 1.5 billion people are afflicted with at least one NTD type. Among NTDs, leishmaniasis, chagas disease (CD) and human African trypanosomiasis (HAT) result in substantial morbidity and death, particularly within impoverished...

The elimination of human African trypanosomiasis: Monitoring progress towards the 2021 –2030 WHO road map targets
by Jose R. Franco, Gerardo Priotto, Massimo Paone, Giuliano Cecchi, Agustin Kadima Ebeja, Pere P. Simarro, Dieudonne Sankara, Samia B. A. Metwally, Daniel Dagne Argaw BackgroundHuman African trypanosomiasis (HAT) is a neglected tropical disease that usually occurs in rural areas in sub-Saharan Africa. It caused devastating epidemics during the 20th century. Sustained, coordinated efforts by different stakeholders working with national sleeping sickness control programmes (NSSCPs) succeeded in controlling the disease and reducing the number of cases to historically low levels. In 2012, WHO targeted the elimination of the di...

Modelled impact of Tiny Targets on the distribution and abundance of riverine tsetse
ConclusionsDespite its simplifications, the model can assist planning and teaching, but allowance should be made for any complications due to seasonality and management challenges associated with greater scale. (Source: PLoS Neglected Tropical Diseases)

< i > Ginkgo biloba < /i > attenuated detrimental inflammatory and oxidative events due to < i > Trypanosoma brucei rhodesiense < /i > in mice treated with melarsoprol
ConclusionUtilization of GB as an adjuvant therapy may ameliorate detrimental effects caused by T.b.r infection and MelB toxicity during late stage HAT. (Source: PLoS Neglected Tropical Diseases)

GSE261736 Depolymerization of SUMO chains induces slender to stumpy differentiation in T. brucei bloodstream parasites
Contributors : Paula A Iribarren ; Luc ía A Di Marzio ; María A Berazategui ; Andreu Saura ; Lorena Coria ; Juliana Cassataro ; Federico Rojas ; Miguel Navarro ; Vanina E AlvarezSeries Type : Expression profiling by high throughput sequencingOrganism : Trypanosoma bruceiTrypanosoma brucei are protozoan parasites that cause sleeping sickness in humans and nagana in cattle. Inside the mammalian host, a quorum sensing-like mechanism coordinates its differentiation from a slender replicative form into a quiescent stumpy form, limiting growth and virulence. SUMOylation is a reversible post-translational modification that...

Molecules, Vol. 29, Pages 1611: Antiprotozoal Activity of Plants Used in the Management of Sleeping Sickness in Angola and Bioactivity-Guided Fractionation of Brasenia schreberi J.F.Gmel and Nymphaea lotus L. Active against T. b. rhodesiense
Wolfender Pascal Mäser Folk medicine is widely used in Angola, even for human African trypanosomiasis (sleeping sickness) in spite of the fact that the reference treatment is available for free. Aiming to validate herbal remedies in use, we selected nine medicinal plants and assessed their antitrypanosomal activity. A total of 122 extracts were prepared using different plant parts and solvents. A total of 15 extracts from seven different plants exhibited in vitro activity (>70% at 20 µg/mL) against Trypanosoma brucei rhodesiense bloodstream forms. The dichloromethane extract of Nymphaea lotu...

Comparison of stochastic and deterministic models for < i > gambiense < /i > sleeping sickness at different spatial scales: A health area analysis in the DRC
We present a previously developed stochastic mathematical model for gHAT in the Democratic Republic of Congo (DRC) and show that this same formulation is able to capture the dynamics of gHAT observed at the health area level (approximately 10,000 people). This analysis was the first time any stochastic gHAT model has been fitted directly to case data and allows us to better quantify the uncertainty in our results. The analysis focuses on utilising a particle filter Markov chain Monte Carlo (MCMC) methodology to fit the model to the data from 16 health areas of Mosango health zone in Kwilu province as a case study. The spat...

Suramin: Effectiveness of analogues reveals structural features that are important for the potent trypanocidal activity of the drug
Exp Parasitol. 2024 Mar 19:108744. doi: 10.1016/j.exppara.2024.108744. Online ahead of print.ABSTRACTSuramin was the first effective drug for the treatment of human African sleeping sickness. Structural analogues of the trypanocide have previously been shown to be potent inhibitors of several enzymes. Therefore, four suramin analogues lacking the methyl group on the intermediate rings and with different regiochemistry of the naphthalenetrisulphonic acid groups and the phenyl rings were tested to establish whether they exhibited improved antiproliferative activity against bloodstream forms of Trypanosomes brucei compared to...

Print 2024 Feb 27.ABSTRACTBACKGROUND: African sleeping sickness is a neglected tropical disease seldom seen in European travellers.CASE PRESENTATION: While working in Eastern Africa, a Norwegian man in his sixties developed weakness and fever. He was prescribed doxycycline after a negative malaria rapid test. On the third day of illness he returned to Norway and was admitted to the hospital upon arrival. On admission he was somnolent with fever, tachypnoea, tachycardia, jaundice, a hyperaemic rash, oliguria and haematuria. Blood tests revealed leukop...
