Low-grade uterine Leiomyosarcoma is highly sensitive to hormonal treatment
CONCLUSIONS: We observed a response or long-term stability rate on hormonal therapy in the 90% range; in all cases the time to relapse was significantly longer than in ULMS and in most cases the relapse was abdominal. Based on these findings, we conclude that a proportion of patients with uterine smooth muscle neoplasms actually present with a "low-grade ULMS".PMID:37699075 | DOI:10.1158/1078-0432.CCR-23-0692 (Source: Clinical Cancer Research)

Pituitary metastases: a case series and scoping review
ConclusionsThis scoping review showed that both radiosurgery and surgical resection have been frequently used to treat pituitary metastases with good response. Vision improvement is more likely to happen following surgical resection, likely at the expense of endocrine dysfunction. Despite treatment and radiographic response, patient survival remains less than a year. (Source: Pituitary)

Aggressive surgical approach with vascular resection and reconstruction for retroperitoneal sarcomas: a systematic review
Surgery is the mainstay of treatment and completeness of surgical resection is critical to achieve local control for retroperitoneal sarcoma (RPS). En-bloc resection of adjacent organs, including major abdomin... (Source: BMC Surgery)

Identification of ARAP3 as a regulator of tumor progression, macrophage infiltration and osteoclast differentiation in a tumor microenvironment-related prognostic model of Ewing sarcoma
In this study, we not only evaluated the gene sets associated with TME in ES using ESTIMATE and WGCNA algorithms based on the transcriptome data of ES, but also constructed a prognostic model (ES Score) using univariate Cox regression and Lasso regression and assessed its predictive ability on immune cell infiltration. Subsequently, we identified ARAP3 as a key gene affecting the TME of ES. In addition, bioinformatic analyses and in vitro experiments proved that the high expression of ARAP3 regulated ES cell proliferation, migration, as well as apoptosis via the p53 signaling pathway and affected macrophage infiltration an...

ATRX loss suppresses the type I interferon response in sarcoma cells through chromatin remodeling
Am J Cancer Res. 2023 Aug 15;13(8):3547-3558. eCollection 2023.ABSTRACTSarcomas constitute a heterogeneous group of mesenchymal cancers and are particularly common in children and adolescents, leading to significant lethality. Therefore, it is necessary to understand the underlying mechanisms by which genetic alterations promote sarcoma progression. Here, we demonstrate that loss-of-function of ATRX, a member of the SWI/SNF DNA-remodeling family, represses the interferon (IFN)-β response by inducing chromatin remodeling in sarcoma cells. We show that ATRX mutations are associated with worse prognosis and attenuate IFN-α/...

Age and treatment disparities in survival of primary malignant cardiac tumors: an analysis of over 40 years and 500 patients
CONCLUSIONS: This study confirms that PMCTs have limited treatment options and poor patient survival, especially for elderly patients and patients who receive no treatment. And patients with PMCTs of any age, whether treated or not, have poor survival rates. Techniques for early diagnosis and treatment may be necessary. Surgical treatment should have a higher priority for future treatment of patients with sarcomas.PMID:37691678 | PMC:PMC10482641 | DOI:10.21037/jtd-23-1054 (Source: Journal of Thoracic Disease)

VISTA expression on cancer-associated endothelium selectively prevents T-cell extravasation
Cancer Immunol Res. 2023 Sep 11. doi: 10.1158/2326-6066.CIR-22-0759. Online ahead of print.ABSTRACTCancers evade T-cell immunity by several mechanisms such as secretion of anti-inflammatory cytokines, downregulation of antigen presentation machinery, upregulation of immune checkpoint molecules, and exclusion of T cells from tumor tissues. The distribution and function of immune checkpoint molecules on tumor cells and tumor-infiltrating leukocytes is well established, but less is known about their impact on intratumoral endothelial cells. Here, we demonstrated that V-domain Ig suppressor of T-cell activation (VISTA), a PD-L...
