Reversal of dual epigenetic repression of non-canonical Wnt-5a normalises diabetic corneal epithelial wound healing and stem cells
Conclusions/interpretationWe provide the first account of epigenetic changes in diabetic corneas including dual inhibition ofWNT5A by DNA methylation and miRNA action. Overall, Wnt-5a is a new corneal epithelial wound healing stimulator that can be targeted to improve wound healing and stem cells in the diabetic cornea.Data availabilityThe DNA methylation dataset is available from the public GEO repository under accession no. GSE229328 (https://www.ncbi.nlm.nih.gov/geo/query/acc.cgi?acc=GSE229328).Graphical Abstract

Aberrant PI3K δ splice isoform as a potential biomarker and novel therapeutic target for endocrine cancers
ConclusionIn summary, our study has suggested a promising potential of utilizing PI3Kδ-S (an oncogenic isoform conferring drug resistance and exempt from PTEN regulation) as a prognostic biomarker and drug target in advanced endocrine cancers.

Glucose-stimulated insulin secretion depends on FFA1 and Gq in neonatal mouse islets
Conclusions/interpretationFFA1 promotes glucose-responsive insulin secretion and functional maturation of newborn islets and is required for adaptive offspring insulin secretion in the face of metabolic challenge, such as parental HFD.Graphical Abstract

The role of proteasome activators PA28 αβ and PA200 in brown adipocyte differentiation and function
DiscussionIn summary, we found no role for Psme1 and Psme4 in brown adipocyte proteostasis, differentiation, or function. These findings contribute to our basic understanding of proteasome biology and the roles of proteasome activators in brown adipocytes.

Periostin acts as a bridge between gestational diabetes mellitus (GDM) and chronic inflammation to modulate insulin resistance by modulating PPAR α/NF-κB/TNF-α signaling pathway
CONCLUSION: POSTN levels were significantly higher in pregnant women with GDM and were associated with chronic inflammation and PPARα expression. POSTN may act as a bridge between GDM and chronic inflammation to modulate insulin resistance by modulating PPARα/NF-κB/TNF-α signaling pathway.PMID:37106515 | DOI:10.2174/1871530323666230427104724

A functional genetic screen for metabolic proteins unveils GART and the de novo purine biosynthetic pathway as novel targets for the treatment of luminal A ER α expressing primary and metastatic invasive ductal carcinoma
In conclusion, GART inhibition by LMX or other inhibitors of the de novo purine biosynthetic pathway could be a novel effective strategy for the treatment of primary and metastatic BCs.

Chaperonin counteracts diet-induced non-alcoholic fatty liver disease by aiding sirtuin 3 in the control of fatty acid oxidation
Conclusion/interpretationMitochondrial HSP60 promotes fatty acid oxidation while repressing mitochondrial stress and inflammation to ameliorate the development of NAFLD by preserving SIRT3 signalling. This study reveals the hepatoprotective effects of HSP60 and indicates that HSP60 could play a fundamental role in the development of therapeutics for NAFLD or type 2 diabetes.Graphical abstract

Role of SUMO-Specific Protease 2 in Leptin-Induced Fatty Acid Metabolism in White Adipocytes
CONCLUSION: These results suggest that the SENP2-PPARα pathway plays an important role in leptin-induced FAO in white adipocytes.PMID:36872063 | DOI:10.4093/dmj.2022.0156
