DAB2IP-knocking down resulted in radio-resistance of breast cancer cells is associated with increased hypoxia and vasculogenic mimicry formation
Conclusions: Our data support that loss of DAB2IP confers radio-resistance of BCa could be due to increased hypoxia, inhibited vascular maturation and promoted VM formation. STAT3 inhibition could be a potential way to overcome such DAB2IP-deficient induced tolerance in BCT.PMID:36947637 | DOI:10.1080/09553002.2023.2194390

OTOR in breast carcinoma as a potent prognostic predictor correlates with cell proliferation, migration and invasion.
This study aimed to investigate clinicopathologic significance of OTOR in breast infiltrating ductal carcinoma (IDC) with high metastasis and unveil its biological function in BC. OTOR was highly overexpressed in BC tissues and cells compared with normal samples. OTOR overexpression was associated with a few of clinicopathologic characteristics and worse overall survival (OS) of IDC breast patients. Silencing of OTOR using siRNA tool impeded BC cells proliferation, migration and invasion by CCK-8, colony formation, wound-healing and transwell assays, which might be regulated by mitogen-activated protein kinase/extracellula...

Abstract P2-11-09: A p53-based strategy for protecting normal breast tissue from chemotherapy-induced damage in breast conserving therapy
Conclusions:Together our data show that the MAPK pathway is hyperactivated in TNBC; inhibiting this pathway impairs tumor growth, but enhances GPNMB, which facilitates mammary tumor growth and metastasis in the setting of Mek-i. These data provide rationale for combined targeting of GPNMB and the MAPK pathway in TNBC.Citation Format: Long M, Huang Y, Liu R, Liu R, Su H. Ionizing Radiation Sensitizes Breast Cancer Cells to Bcl-2 Inhibitor, ABT-737, through Regulating Mcl-1.
Abstract Breast-conserving surgery followed by radiation therapy has become the standard of care for early stage breast cancer. However, there are some patients that develop a local failure. We have previously shown that Bcl-2 overexpression was associated with an increased risk of local recurrence in patients with early stage breast cancer. The purpose of this study was to explore an approach to overcome radiation resistance by targeting pro-survival Bcl-2 family proteins in breast cancer cells. The breast cancer cell lines MCF-7, ZR-75-1 and MDA-MB231 were used in this study. siRNAs were employed to silence myel...
