Overcoming the non-kinetic activity of EGFR1 using multi-functionalized mesoporous silica nanocarrier for in vitro delivery of siRNA
Sci Rep. 2022 Oct 14;12(1):17208. doi: 10.1038/s41598-022-21601-w.ABSTRACTTriple-negative breast cancer (TNBC) does not respond to HER2-targeted and hormone-based medicines. Epidermal growth factor receptor 1 (EGFR1) is commonly overexpressed in up to 70% of TNBC cases, so targeting cancer cells via this receptor could emerge as a favored modality for TNBC therapy due to its target specificity. The development of mesoporous silica nanoparticles (MSNs) as carriers for siRNAs remains a rapidly growing area of research. For this purpose, a multi-functionalized KIT-6 containing the guanidinium ionic liquid (GuIL), PEI and PEGy...

Crosstalk between TBK1/IKK ε and the type 1 interferon pathway contributes to tubulointerstitial inflammation and kidney tubular injury
In conclusion, T1-IFNs, TWEAK, and LPS engage interrelated proinflammatory and antiviral responses in tubular cells. Moreover, inhibition of TBK1/IKKε with amlexanox, and IFNAR targeting, may protect from tubulointerstitial kidney injury.

A cationic amino acid polymer nanocarrier synthesized in supercritical CO < sub > 2 < /sub > for co-delivery of drug and gene to cervical cancer cells
Colloids Surf B Biointerfaces. 2022 May 18;216:112584. doi: 10.1016/j.colsurfb.2022.112584. Online ahead of print.ABSTRACTThe present study was undertaken to investigate the ability of a drug curcumin-loaded polymer to inhibit the growth of cervical cancer cells by enhancing the anti-cancer efficiency of curcumin. We synthesized poly(methacryloyl beta-alanine) (PMBA) as a nanocarrier by radical polymerization in supercritical CO2. The results showed that the curcumin encapsulated and folic acid (FA)-treated PMBA (Poly@Cur-FA) for 24 h activated the reactive oxygen species-mediated programmed cell death machinery in HeLa ce...

Microencapsulated Multifunctionalized Graphene Oxide Equipped with Chloroquine for Efficient and Sustained siRNA Delivery
Biomed Res Int. 2022 Apr 6;2022:5866361. doi: 10.1155/2022/5866361. eCollection 2022.ABSTRACTA multifunctionalized graphene oxide (GO)-based carrier with conjugation of aminated-polyethylene glycol (PEG-diamine), octaarginine (R8), and folic acid (FA), which also contains chloroquine (CQ), a lysosomotropic agent, is introduced. The cellular uptake mechanisms and intracellular targeting of FA-functionalized nanocarriers are examined. The localized releases of CQ and siRNA intracellular delivery are evaluated. Microencapsulation of the nanocarrier complexed with genes in layer-by-layer coating of alginate microbeads is also ...

Simultaneous blockade of TIGIT and HIF-1 α induces synergistic anti-tumor effect and decreases the growth and development of cancer cells
CONCLUSIONS: Present data suggest the combination treatment of TIGIT and HIF-1α as a novel treatment strategy against colorectal and breast cancer, but more researches are required to realize the complete role of TIGIT and HIF-1α inside the TME.PMID:34710844 | DOI:10.1016/j.intimp.2021.108288
