Melatonin Postconditioning Ameliorates Anoxia/Reoxygenation Injury by Regulating Mitophagy and Mitochondrial Dynamics in a SIRT3-Dependent Manner
Eur J Pharmacol. 2021 May 7:174157. doi: 10.1016/j.ejphar.2021.174157. Online ahead of print.ABSTRACTIschaemia/reperfusion (I/R) injury is accompanied by excessive mitochondrial autophagy (mitophagy) and an imbalance in mitochondrial dynamics. Melatonin has been reported to alleviate I/R injury by regulating mitophagy and mitochondrial dynamics. However, the underlying mechanism associated with this activity is not fully understood. The goal of the present study was to investigate whether and how melatonin administration at the beginning of reoxygenation exerts protective effects by regulating mitophagy and mitochondrial d...

Mechanistic study of mtROS-JNK-SOD2 signaling in bupivacaine-induced neuron oxidative stress.
Abstract Manganese superoxide dismutase (SOD2) is a key enzyme to scavenge free radical superoxide in the mitochondrion. SOD2 deficiency leads to oxidative injury in cells. Bupivacaine, a local anesthetic commonly used in clinic, could induce neurotoxic injury via oxidative stress. The role and the mechanism of SOD2 regulation in bupivacaine-induced oxidative stress remains unclear. Here, bupivacaine was used to treat Sprague-Dawley rats with intrathecal injection and culture human neuroblastoma cells for developing vivo injury model and vitro injury model. The results showed that bupivacaine caused the over-produ...

The solute carriers ZIP8 and ZIP14 regulate manganese accumulation in brain microvascular endothelial cells and control brain manganese levels Neurobiology
In this study, we have determined that ZIP8 and ZIP14 (Zrt- and Irt-like proteins 8 and 14) support Mn2+ uptake by BMVECs and that neither DMT1 nor an endocytosis-dependent pathway play any significant role in Mn2+ uptake. Specifically, siRNA-mediated knockdown of ZIP8 and ZIP14 coincided with a decrease in manganese uptake, and kinetic analyses revealed that manganese uptake depends on pH and bicarbonate and is up-regulated by lipopolysaccharide, all biochemical markers of ZIP8 or ZIP14 activity. Mn2+ uptake also was associated with cell-surface membrane presentation of ZIP8 and ZIP14, as indicated by membrane protein bio...

Activatable MRI-monitoring gene delivery for the theranostic of renal carcinoma
In this study, we constructed a PEG-MnO2-OPN siRNA nanocomplex via a modular streptavidin bridge for the theranostic treatment of renal carcinoma in vitro and in vivo. This strategy of utilizing PEG-MnO2 nanosheets for OPN siRNA delivery showed effective tumor growth inhibition of 786-O tumor-bearing mice, and such a process could be monitored by the activated T1-weight MRI, illustrating the promising potential of PEG-MnO2 nanosheets for the fabrication of an MRI-based theranostic system.

Vitamin C Attenuates Sodium Fluoride-Induced Mitochondrial Oxidative Stress and Apoptosis via Sirt1-SOD2 Pathway in F9 Cells.
Abstract Increasing evidence has suggested an important role played by reactive oxygen species (ROS) in the pathogenesis of fluorosis. Accumulating evidence demonstrates that vitamin C administration ameliorate sodium fluoride (NaF)-induced oxidative stress. However, the potentially beneficial effects of vitamin C against NaF-induced cytotoxicity and the underlying molecular mechanisms of this protection are not fully understood. Here, we found that NaF stimulated cytotoxicity, increased mitochondrial reactive oxygen species (mROS) production, and induced apoptosis in F9 embryonic carcinoma cells. Consistent with ...

Melatonin protects against myocardial ischemia-reperfusion injury by elevating Sirtuin3 expression and manganese superoxide dismutase activity.
Authors: Feng J, Chen X, Liu R, Cao C, Zhang W, Zhao Y, Nie S Abstract Myocardial ischemia-reperfusion (MI/R) injury is a crucial cause for mortality throughout the world. Recent studies indicated that melatonin might exert profound cardio-protective effect in MI/R injury. However, the underlying mechanisms are not completely understood. In the current study, we aimed to explore the potential effect of melatonin in the pathological process of MI/R. Both in vivo MI/R model and in vitro H9c2 cell line simulated I/R (SIR) model were applied with or without melatonin supplementation. We found that Sirtuin3 (Sirt3) expr...

Bush sophora root polysaccharide could help prevent aflatoxin B1-induced hepatotoxicity in the primary chicken hepatocytes.
Abstract The aim of this study was to evaluate the effects of bush sophora root polysaccharide (BSRPS) on the aflatoxin B1 (AFB1)-induced hepatotoxicity and to explore the underlying mechanisms. The primary chicken hepatocytes were used as the model in the present experiment. The results showed that AFB1 induced hepatotoxicity of chicken hepatocytes in a dose dependent manner as demonstrated by decreasing cell viability and increasing LDH activity, ALT and AST levels. AFB1 at 0.16 μM significantly increased the levels of hepatic cytochrome P450 1A5 (CYP450 1A5) mRNA and malondialdehyde (MDA) and decreased the a...

MicroRNA-140-5p attenuated oxidative stress in Cisplatin induced Acute Kidney Injury by activating Nrf2/ARE pathway through a Keap1-independent mechanism.
Abstract Oxidative stress was predominantly involved in the pathogenesis of acute kidney injury (AKI). Recent studies had reported the protective role of specific microRNAs (miRNAs) against oxidative stress. Hence, we investigated the levels of miR140-5p and its functional role in the pathogenesis of Cisplatin induced AKI. A mice Cisplatin induced-AKI model was established. We found that miR-140-5p expression was markedly increased in mice kidney. Bioinformatics analysis revealed nuclear factor erythroid 2-related factor (Nrf2) was a potential target of miR-140-5p, We demonstrated that miR-140-5p did not affect Ke...

Resveratrol alleviates diabetic cardiomyopathy in rats by improving mitochondrial function through PGC-1 α deacetylation.
In this study we examined whether resveratrol alleviated cardiac dysfunction in diabetes by improving mitochondrial function via SIRT1-mediated PGC-1α deacetylation. T2DM was induced in rats by a high-fat diet combined with STZ injection. Diabetic rats were orally administered RSV (50 mg·kg(-1)·d(-1)) for 16 weeks. RSV administration significantly attenuated diabetes-induced cardiac dysfunction and hypertrophy evidenced by increasing ejection fraction (EF%), fraction shortening (FS%), ratio of early diastolic peak velocity (E velocity) and late diastolic peak velocity (A velocity) of the LV inflow (E/A ratio) and reduci...

MnAl-Layered Double Hydroxide Nanoparticles as a Dual-functional Platform for Magnetic Resonance Imaging and siRNA Delivery.
Abstract Multifunctional nanoparticles for cancer theranosis have been widely explored for effective cancer detection and therapy. In this research, dually functionalised manganese-based layered double hydroxide nanoparticles (Mn-LDHs) have been examined as an effective anticancer drug/gene delivery system and T1-weighted magnetic resonance imaging (MRI) modality for brain cancer theranostics. Such Mn-LDHs have been shown to accommodate dsDNA/siRNAs and efficiently deliver to Neuro-2a cells (N2a). Mn-LDHs have also shown high biocompatibility with low cytotoxicity. Importantly, the cell death-siRNA (CD-siRNA) deli...
