Folate receptor-targeted RNAi nanoparticles for silencing STAT3 in tumor-associated macrophages and tumor cells
Publication date: Available online 18 February 2020Source: Nanomedicine: Nanotechnology, Biology and MedicineAuthor(s): Jing Chen, Yushun Dou, Yue Tang, Xinru ZhangAbstractWe developed a STAT3 silencing siRNA to both tumor cells and M2 macrophages. The dual-targeting system prepared by electronic self-assembly was composed of folic acid-conjugated carboxymethyl chitosan for targeting and cationic chitosan derivatives for siRNA package. The effects of siRNA delivery was investigated in M2 macropahges and Lewis lung cancer cells(LLC). Due to the enhanced delivery efficiency, the dual-targeting delivery system exhibited a hig...

Enriched chitosan nanoparticles loaded with siRNA are effective in lowering Huntington's disease gene expression following intranasal administration
Publication date: Available online 27 October 2019Source: Nanomedicine: Nanotechnology, Biology and MedicineAuthor(s): Vasyl Sava, Oksana Fihurka, Anastasia Khvorova, Juan Sanchez-RamosAbstractTherapies to lower gene expression in brain disease currently require chronic administration into the cerebrospinal fluid (CSF) by intrathecal infusions or direct intracerebral injections. Though well-tolerated in the short-term, this approach is not tenable for a life-time of administration. Nose-to-brain delivery of enriched chitosan-based nanoparticles loaded with anti-HTT siRNA was studied in a transgenic YAC128 mouse model of Hu...

Site-specific intestinal DMT1 silencing to mitigate iron absorption using pH-sensitive multi-compartmental nanoparticulate oral delivery system
We report a novel pH-sensitive multi-compartmental nanoparticulate (MCP) oral delivery system that encapsulates siRNA for silencing the divalent metal transporter 1 (DMT1) gene, a primary iron transporter highly expressed in the intestine. The gelatin nanoparticles-coated with Eudragit® L100–55 are able to protect siRNA from intracellular degradation and allow for sustained release of siRNA in the cytosol. When orally-administered, Eudragit® L100–55 based MCPs containing DMT1 siRNA/gelatin NPs specifically and significantly decreased the expression of DMT1 mRNA in the duodenum, which consequently decreased intestinal...

Single-Cell Kinetics of siRNA-Mediated mRNA Degradation
Publication date: Available online 8 August 2019Source: Nanomedicine: Nanotechnology, Biology and MedicineAuthor(s): Rafał Krzysztoń, Daniel Woschée, Anita Reiser, Gerlinde Schwake, Helmut H. Strey, Joachim O. RädlerAbstractRNA interference (RNAi) enables the therapeutic use of small interfering RNAs (siRNAs) to silence disease-related genes. The efficiency of silencing is commonly assessed by measuring expression levels of the target protein at a given time point post-transfection. Here, we determine the siRNA-induced fold change in mRNA degradation kinetics from single-cell fluorescence time-courses obtained using li...

Delivery of siRNA using folate receptor-targeted pH-sensitive polymeric nanoparticles for rheumatoid arthritis therapy
Publication date: Available online 22 May 2019Source: Nanomedicine: Nanotechnology, Biology and MedicineAuthor(s): Xiangshi Sun, Shiyan Dong, Xiangyu Li, Kongtong Yu, Fengying Sun, Robert J. Lee, Youxin Li, Lesheng TengAbstractSystemic delivery of siRNA to target tissues is difficult to achieve owing to its limited cellular uptake and poor serum stability. Herein, polymeric nanoparticles were developed for systemic administration of siRNA to inflamed tissues. The polymeric nanoparticles were composed of PK3 as a pH-sensitive polymer, folate-polyethyleneglycol-poly(lactide-co-glycolide) as a targeting ligand, and a DOTAP/si...
