1173 Inducible DNA repair of UV photoproducts depends on vitamin D receptor
Ultraviolet radiation (UV) initiates vitamin D synthesis by converting 7-dehydrocholesterol to pre-vitamin D3 that is converted to 25-hydroxyvitamin D3 (25D3) and then 1,25-dihydroxyvitamin D3 (1,25D3). However, UV also generates DNA damage that is repaired by nucleotide excision repair (NER). We tested the hypothesis that vitamin D signaling elicits compensatory responses to the DNA damage incurred during vitamin D synthesis. Ketoconazole induces apoptosis in rat cardiomyocytes through reactive oxygen species-mediated parkin overexpression.
Abstract Azole antifungals such as ketoconazole are generally known to induce a variety of heart function side effects, e.g., long-QT syndrome and ventricular arrhythmias. However, a clear mechanism for the action of ketoconazole in heart cells has not been reported. In the present study, we assessed the correlation between ketoconazole-induced apoptosis and the alteration of genes in response to ketoconazole in rat cardiomyocytes. Abstract 4751: 25-hydroxyvitamin D3 induces vitamin D signaling independent of CYP27B1 in non-small cell lung cancer cells
Conclusions NSCLC cells expressing high levels of VDR display increased responsiveness to vitamin D metabolites. Although NSCLC cells express CYP27B1, our results demonstrate that the enzyme may not be required to achieve vitamin D signaling. Rather, 25(OH)D3 may act via the VDR to elicit an anti-tumor responses. The implication of these findings is that dietary supplementation to increase circulating 25(OH)D3 may be beneficial in a subset of NSCLC patients.
