E-258 Endovascular approach to drug delivery to the brain
ConclusionsCPA delivery of AAV9 resulted in increased transduction of the parietal and cingulate cortex, comparable to that observed with CM injections. These results provide preliminary evidence that CPA infusion of gene therapy, and potentially other therapies such as antisense oligonucleotides or siRNA medications, is safe and provides widespread distribution throughout the brain.Abstract E-258 Figure 1Disclosures A. The long and short non-coding RNAs modulating EZH2 signaling in cancer
J Hematol Oncol. 2022 Mar 2;15(1):18. doi: 10.1186/s13045-022-01235-1.ABSTRACTNon-coding RNAs (ncRNAs) are a large family of RNA molecules with no capability in encoding proteins. However, they participate in developmental and biological processes and their abnormal expression affects cancer progression. These RNA molecules can function as upstream mediators of different signaling pathways and enhancer of zeste homolog 2 (EZH2) is among them. Briefly, EZH2 belongs to PRCs family and can exert functional roles in cells due to its methyltransferase activity. EZH2 affects gene expression via inducing H3K27me3. In the present ...

Bioinformatics analyses show dysregulation of calcium-related genes in Angelman syndrome mouse model.
Abstract BACKGROUND: Angelman syndrome (AS) is a genetic neurodevelopmental disorder caused by the loss of function of the UBE3A protein in the brain. In a previous study, we showed that activity-dependent calcium dynamics in hippocampal CA1 pyramidal neurons of AS mice is compromised, and its normalization rescues the hippocampal-dependent deficits. Therefore, we expected that the expression profiles of calcium-related genes would be altered in AS mice hippocampi. METHODS: We analyzed mRNA sequencing data from AS model mice and WT controls in light of the newly published CaGeDB database of calcium-related ge...

Oligonucleotides —A Novel Promising Therapeutic Option for IBD
Conclusions In this review, we focused on recent and past approaches to test the therapeutic efficacy of oligonucleotide based therapies in IBD. The combining mechanistic mode of oligonucleotide based therapeutics is a targeted action on specific pro-inflammatory molecules, which are over activated in IBD patients and contribute significantly to disease pathogenesis. The proposed high selectivity of the agents is derived from its mode of action, that aims to specifically block certain inflammatory molecular patterns, without a general systemic effect on other molecular targets. It would be important for each oligonucleot...

Tumor-Stroma-Inflammation Networks Promote Pro-metastatic Chemokines and Aggressiveness Characteristics in Triple-Negative Breast Cancer
In this study, MSCs of four different healthy donors were used. Patient-derived CAFs from a primary breast tumor (used in ELISA and their accompanying signaling experiments) and from a lung metastasis (used in tumor cell invasion assays) were kindly provided by Dr. Bar, Sheba Medical Center, Ramat Gan, Israel). The cells were grown, identified and immortalized as described in Katanov et al. (67). TNFα and IL-1β Concentrations Used in Different Analyses Titration studies were initiated by determining the ability of rhTNFα (#300-01A, PeproTech, Rocky Hill, NJ), and rhIL-1β (#...

Complement C5b-9 and Cancer: Mechanisms of Cell Damage, Cancer Counteractions, and Approaches for Intervention
In conclusion, osmotic burst of inflated complement-damaged cells may occur, but these bursts are most likely a consequence of metabolic collapse of the cell rather than the cause of cell death. The Complement Cell Death Mediator: A Concerted Action of Toxic Moieties Membrane pores caused by complement were first visualized by electron microscopy on red blood cell membranes as large ring structures (22). Similar lesions were viewed on E. coli cell walls (23). Over the years, ample information on the fine ultrastructure of the MAC that can activate cell death has been gathered (24) and has been recently further examined (...

The role of a single gene encoding the Single von Willebrand factor C-domain protein (SVC) in bumblebee immunity extends beyond antiviral defense.
Abstract The Single von Willebrand factor C-domain proteins (SVCs) are a group of short proteins mainly found in arthropods. They are proposed to be responsive in relation to environmental challenges including the nutritional status, bacterial and viral infections. The SVC protein Vago acts as a cross-talk molecule between the small interfering RNA (siRNA) pathway and the Jak/STAT pathway upon viral infection in Drosophila melanogaster and Culex mosquito cells. Unlike flies and mosquitoes that possess diverse SVCs, most bee species only have one of which the function remains unclear. Here we investigated whether t...
