Daphnepedunins G and H, anti-HIV macrocyclic 3,4-seco-daphnane orthoesters from Daphne pedunculata
AbstractDaphnepedunins G (1) and H (2) with unusual macrocyclic 3,4-seco-daphnane orthoester structure were isolated fromDaphne pedunculata. Their structures were determined by physicochemical and spectroscopic analyses combined with synthetic methods, including methyl esterification, derivatization reaction using a chiral anisotropic agent, and biomimetic conversion. Compounds1 and2 along with their methyl esters1a and2a were evaluated for anti-HIV activity, among which1a and2a exhibited potent activity with IC50 values of 1.08 and 1.17  μM, respectively.Graphical abstract

In this study, TTV replication was evaluated in 21 people with acute HIV infection (AHI) and immune reconstitution following antiretroviral therapy (ART). PBMC-associated TTV and HIV-1 DNA, as well as plasma HIV-1 RNA, were measured by real-time PCR. CD4 and CD8 differentiation, activation, exhaustion, and senescence phenotypes were analyzed by flow cytometry. Thirteen healthy donors (HD) and twenty-eight chronically infected HIV individuals (CHI), late presenters at diagnosis, were included as control groups. ConclusionsIn conclusion, the results show that HIV risk behaviours are prevalent among the married and cohabiting population in Botswana. Therefore, policies and programmes relating to issues such as gender inequality and alcohol consumption should be target-specific to address the married and cohabiting individuals.
