Glioma Response to IDH Inhibition: Real-World Experience
Clin Cancer Res. 2023 Sep 22. doi: 10.1158/1078-0432.CCR-23-2164. Online ahead of print.ABSTRACTTreatment of IDH mutated non-enhancing grade 2 and 3 diffuse gliomas with ivosidenib leads to reduction of tumor size when assessed via volumetric MRI. IDH inhibition has a therapeutic benefit in patients with these tumors.PMID:37738033 | DOI:10.1158/1078-0432.CCR-23-2164

Genetic Predictors of Ibrutinib-Related Cardiovascular Side Effects in Patients with Chronic Lymphocytic Leukemia
CONCLUSIONS: Our findings suggest possible genetic determinants of ibrutinib-related CVSEs in CLL. If replicated in a larger study, pre-treatment pharmacogenetic testing for GATA4 and KCNQ1 polymorphisms may be a useful clinical tool for personalizing treatment selection for CLL and/or instituting early risk-mitigation strategies.PMID:37738027 | DOI:10.1158/1078-0432.CCR-23-0421

Depletion of mTOR ameliorates CD < sub > 4 < /sub > < sup > + < /sup > T cell pyroptosis by promoting autophagy activity in septic mice
In this study, a septic mouse model was developed using cecal ligation and puncture (CLP) to explore the degree of pyroptosis and autophagy of CD4+ T cells. T-cell-specific mTOR/TSC1-knockout mice were used to investigate the role of mTOR pathway in the regulation of CD4+ T cell pyroptosis. Bafilomycin, a specific autophagy inhibitor, was used to verify the regulatory effect of autophagy on pyroptosis in septic mice. We observed aggravated pyroptosis in CD4+ T cells in CLP mice accompanied by impaired autophagy activity and an overactivated mTOR signaling pathway. Depletion of mTOR relieved autophagy deficiency and reduced...

Meta-analysis of primary care delivered buprenorphine treatment retention outcomes
Conclusions: While we only investigate BT through primary care, our findings indicate retention rates are equivalent to the rates reported in the specialty care literature. More work is needed to examine factors that may impact primary care delivered BT specifically and differentiate participants that may benefit from care delivered in specialty over primary care as well as the converse.PMID:37737714 | DOI:10.1080/00952990.2023.2251653

The Lysosomal Calcium Channel TRPML1 Maintains Mitochondrial Fitness in NK Cells through Interorganelle Cross-Talk
In this study, we examined the role of TRPML1 (TRP cation channel, mucolipin subfamily, member 1) in regulating the homeostasis of secretory lysosomes and their cross-talk with mitochondria in human NK cells. We found that genetic deletion of TRPML1, which localizes to lysosomes in NK cells, led to mitochondrial fragmentation with evidence of collapsed mitochondrial cristae. Consequently, TRPML1-/- NK92 (NK92ML1-/-) displayed loss of mitochondrial membrane potential, increased reactive oxygen species stress, reduced ATP production, and compromised respiratory capacity. Using sensitive organelle-specific probes, we observed...

Mass Spectrometric Profiling of HLA-B44 Peptidomes Provides Evidence for Tapasin-Mediated Tryptophan Editing
J Immunol. 2023 Sep 22:ji2300232. doi: 10.4049/jimmunol.2300232. Online ahead of print.ABSTRACTThe extreme polymorphisms of HLA class I proteins result in structural variations in their peptide binding sites to achieve diversity in Ag presentation. External factors could independently constrict or alter HLA class I peptide repertoires. Such effects of the assembly factor tapasin were assessed for HLA-B*44:05 (Y116) and a close variant, HLA-B*44:02 (D116), which have low and high tapasin dependence, respectively, for their cell surface expression. Analyses of the HLA-B*44:05 peptidomes in the presence and absence of tapasin...

Opioid Prescribing Habits of Emergency Department Providers in Response to an Educational Intervention
CONCLUSION: Overall, the educational intervention on the topic of opioid prescribing guidelines did not have the expected impact on amounts of opioids administered or prescribed during the study period. Total opioids utilized and opioids prescribed did not vary between the pre- and post-intervention groups overall, by subset diagnosis, or by provider. There was, however, an increase in utilization of non-opioid treatments that could be indicative of a partial response to the educational intervention. This study showed that long-term effects of education regarding recommended opioid utilization might be less effective than ...

Patients with fibromyalgia in Finnish healthcare center - one-year follow up
CONCLUSION: With these results, we cannot conclude that ÖMPSQ predicts the patient's severity of symptoms. On the contrary, we can conclude that antidepressants and/or psychological therapy might reduce fibromyalgia symptoms for depressed patients with fibromyalgia. Patients with fibromyalgia might benefit from clinical evaluation and modifications to the treatment plan if necessary.PMID:37738389 | DOI:10.1080/02813432.2023.2259960

Genetic Disorders in Pediatric Rheumatology Clinic: When to Suspect, and Why?
Indian J Pediatr. 2023 Sep 22. doi: 10.1007/s12098-023-04845-w. Online ahead of print.ABSTRACTJust under a decade ago, most children with genetic disorders received a phenotypic diagnosis, often by atlas matching. With advances in genomics (decoding of human genome, easy availability of genetic testing, and reduction in cost of tests), genotypic diagnosis is now a reality. Genetic diseases can lead to non-inflammatory arthritis that can mimic juvenile idiopathic arthritis (JIA). A small but growing number (as newer genes are discovered) of genetic diseases are being diagnosed in children with a seemingly inflammatory muscu...

Gender-specific results of the Dresden children and adolescents headache program DreKiP
We examined gender differences in the outpatient multimodal Dresden Child and Adolescent Headache Program DreKiP.METHODS: We treated 140 patients with primary headache in a 15-hour structured group program. At baseline (T0) and six (T1) and twelve months (T2) after the end of the program, data on headache-related limitation of daily activities (PedMIDAS) as well as headache frequency, intensity, and pain-related disability (P-PDI) were collected. Retrospectively, these data were analyzed separately for girls and boys.RESULTS: For 91 patients (9-19 years, median = 15; 71.4 % female) data were available for at least two meas...
