ConclusionsFurther work is needed to compare treatment of congenital TTP with PI versus recombinant ADAMTS ‐13. PI may provide longer duration of ADAMTS‐13 effect, but is limited by plasma volume required, whereas recombinant therapy can provide a higher ADAMTS‐13 peak. We propose that investigation of interindividual clearance of ADAMTS‐13 is necessary to optimise treatment, to enable rationale for dose and frequency of prophylaxis.This article is protected by copyright. ConclusionsTF isoforms are expressed in pNET. Sapanisertib suppresses TF mRNA and protein expression as well as TF co ‐factor activityin vitro andin vivo. Ticagrelor blocks the enhanced reactivity of rs773902 A platelets. PAR4 encoded by the rs773902 A allele is relatively resistant to desensitization and may contribute to stroke risk. Natural killer cells induce neutrophil extracellular trap formation in venous thrombosis
Conclusions: Thrombus formation was reduced in Tbet‐/‐ and IFN γ‐/‐ mice, suggesting that Tbet/IFN γ expressing cells are required for venous thrombosis. The number of NETs formed during thrombosis was significantly lower in Tbet‐/‐ and IFN γ‐/‐ mice. NET formation was also decreased in WT mice treated with an IFN γ blocking antibody. Injection of recombinant IFNγ in IFNγ‐/‐ mice rescued the phenotype. Natural killer (NK) cells were specifically depleted prior to venous thrombosis induction. Dimeric Fc γR ectodomains detect pathogenic anti‐platelet factor 4–heparin antibodies in heparin‐induced thromobocytopenia
ConclusionsThis study found the cell ‐free, function‐based dimeric rsFcγRIIa assay to be convenient, simple, and potentially predictive of HIT. The assay had improved specificity over the IgG ELISA, and correlated strongly with the AcuStar HIT IgG‐specific assay, warranting further evaluation of its potential to identify HIT in larger patient cohorts. Safety of D ‐dimer testing as a stand‐alone test for the exclusion of deep vein thrombosis as compared with other strategies
ConclusionsThis outcome study shows that a negative high ‐sensitivity D‐dimer result safely excludes DVT in an outpatient population, and necessitates fewer CUS than if used in combination with Wells score. Increased urokinase and consumption of α2‐antiplasmin as an explanation for the loss of benefit of tranexamic acid after treatment delay
ConclusionsTXA protects fibrin but stimulates uPA activity and slows inhibition of plasmin by α2‐antiplasmin. Plasmin proteolytic activity digests fibrinogen and disrupts coagulation, exacerbated when α2‐antiplasmin is consumed by ongoing fibrinolysis. Additional direct inhibition of plasmin by aprotinin may prevent development of coagulopathy and extend the useful time window of TXA treatment.This article is protected by copyright. Intracranial hemorrhage with direct oral anticoagulants in patients with brain tumors
ConclusionsA total of 172 patients with brain tumors were evaluated (42 DOAC and 131 LMWH). In the primary brain tumor cohort (N=67), the cumulative incidence ofanyICH was 0% in patients receiving DOACs versus 36.8% (95% CI, 22.3 ‐51.3%) in those treated with LMWH with amajorICH incidence of 18.2% (95% CI, 8.4 ‐31.0).
