Potential impact of underlying diseases influencing ADME in nonclinical safety assessment
Food Chem Toxicol. 2024 Apr 4:114636. doi: 10.1016/j.fct.2024.114636. Online ahead of print.ABSTRACTNonclinical studies involve in vitro, in silico, and in vivo experiments to assess the toxicokinetics, toxicology, and safety pharmacology of drugs according to regulatory requirements by a national or international authority. In this review, we summarize the potential effects of various underlying diseases governing the absorption, distribution, metabolism, and excretion (ADME) of drugs to consider the use of animal models of diseases in nonclinical trials. Obesity models showed alterations in hepatic metabolizing enzymes, ...

Identification of herbal drug extracts that promote growth of Akkermansia muciniphila in high-fat diet fed mice
In this study, we screened herbal drug extracts that promoted the growth ofA.muciniphila. Among the 123 herbal drugs, five herbal drug extracts significantly increasedA.muciniphila DNA levels compared with that in controls. In particular, Dioscoreae rhizoma extract increased the growth ofA.muciniphila in the intestines of mice fed a high-fat diet and improved obesity. It significantly reduced body weight gain, improved glucose tolerance even when the administration was initiated after the induction of dietary obesity. These results suggest that herbal drug extracts, such as Dioscoreae rhizome, that increaseA.muciniphila co...

Update in lean metabolic dysfunction-associated steatotic liver disease
CONCLUSION: MASLD is a complex disease comprising epigenetic, genetic, and metabolic factors in its pathogenesis. Results vary across populations, gender, and age. Limited data exists on clinical practice guidelines for lean patients. Future studies employing this new nomenclature can contribute to standardizing and generalizing results among lean patients with steatotic liver disease.PMID:38577539 | PMC:PMC10989317 | DOI:10.4254/wjh.v16.i3.452 (Source: World Journal of Hepatology)

Cardiovascular-Kidney-Metabolic Syndrome: Association with Adverse Events After Major Noncardiac Surgery
CONCLUSION: The newly defined CKM syndrome is associated with increased morbidity and mortality after non-cardiac surgery. Thus, cardiovascular, renal, and metabolic disorders should be regarded in mutual context in this setting.PMID:38578871 | DOI:10.1213/ANE.0000000000006975 (Source: Anesthesia and Analgesia)

Obesity: An Impact with Cardiovascular and Cerebrovascular Diseases
Indian J Clin Biochem. 2024 Apr;39(2):168-178. doi: 10.1007/s12291-023-01157-w. Epub 2023 Oct 13.ABSTRACTThe authors sought to correlate the complex sequel of obesity with various parameters known to develop metabolic syndrome viz. insulin resistance, dyslipidemia, hypertension etc., as these anomalies are linked to vascular atherosclerosis and outbreak of cardiovascular and cerebrovascular diseases. A comprehensive online survey using MEDLINE, Scopus, PubMed and Google Scholar was conducted for relevant journals from 1970 till present time (2023) with key search terms like: 'obesity', 'leptin', type-2 diabetes', 'atherosc...
