Evaluating the rate of reversal of fentanyl-induced respiratory depression using a novel long-acting naloxone nanoparticle, cNLX-NP
DiscussionWhile cNLX-NP alone may not sufficiently reverse F/FA overdose in a timely manner, mixing free NLX with cNLX-NP can provide a mechanism to both rapidly reverse fentanyl-related effects and maintain extended protection against synthetic opioid toxicity. These data support further development of cNLX-NP as a fast-acting and long-lasting antidote to treat F/FA-induced respiratory depression and overdose, and potentially prevent renarcotization in humans. Model-based exploration of the rationality of off-label use of cetirizine in Chinese pediatric patients: a prospective cohort study
Conclusion: This study provides evidence-based and model-based exploration for optimizing cetirizine usage in Chinese pediatric patients. The cetirizine PPK model showed accurate predictive performance and could be utilized to simulate individual patient exposure in real-world clinical scenarios. (Source: Frontiers in Pharmacology)

Clinical Pharmacokinetics and Pharmacodynamics of Naloxone
AbstractNaloxone is a World Health Organization (WHO)-listed essential medicine and is the first choice for treating the respiratory depression of opioids, also by lay-people witnessing an opioid overdose. Naloxone acts by competitive displacement of opioid agonists at the μ-opioid receptor (MOR). Its effect depends on pharmacological characteristics of the opioid agonist, such as dissociation rate from the MOR receptor and constitution of the victim. Aim of treatment is a balancing act between restoration of respiration (not consciousness) and avoidance of withdrawa l, achieved by titration to response after initial dose...

Funding and Delivery of Syringe Services Programs in the United States, 2022
Conclusions. Current SSP funding levels do not meet minimum benchmarks. Increased funding would help SSPs meet community health needs. Public Health Implications. Federal, state, and local initiatives should prioritize sustained SSP funding to optimize their potential in addressing multiple public health crises. Implementation of pharmaceutical alternatives to a toxic drug supply in British Columbia: A mixed methods study
North America has been in an unrelenting overdose crisis for almost a decade. British Columbia (BC), Canada declared a public health emergency due to overdoses in 2016. Risk Mitigation Guidance (RMG) for prescribing pharmaceutical opioids, stimulants and benzodiazepine alternatives to the toxic drug supply ( “safer supply”) was implemented in March 2020 in an attempt to reduce harms of COVID-19 and overdose deaths in BC during dual declared public health emergencies. Our objective was to describe early implementation of RMG among prescribers in BC. (Source: Journal of Substance Abuse Treatment)

Many 12th Graders Found to Use Potentially Risky Products With “Legal” THC Variant
Astudy published today inJAMA reports that many high school seniors are using products containing Δ8-tetrahydrocannabinol (THC), a variant of the main psychoactive compound of marijuana, Δ9-THC. The analysis also showed that Δ8-THC use was elevated in states where recreational marijuana was illegal.“Gummies and other edibles, electronic vaping devices, and combustible flower containing Δ8-THC are marketed as providing a user experience comparable to marijuana in a product that is federally legal,” wrote Alyssa F. Harlow, Ph.D., of the University of Southern California and colleagues. “Δ 8-THC exposure may pose r...

Targeting gut microbiota to counteract acetaminophen-induced acute liver injury
Trends Microbiol. 2024 Mar 11:S0966-842X(24)00051-9. doi: 10.1016/j.tim.2024.02.011. Online ahead of print.ABSTRACTAcetaminophen (N-acetyl-p-aminophenol; APAP) overdose-induced acute liver injury (AILI) is a huge threat to public health worldwide. Recent research clearly shows that the intestinal microbiota (IM) is a key modulator in AILI. Herein, I discuss the latest findings on how the IM regulates AILI and the potential interventions to combat AILI by targeting the IM.PMID:38472076 | DOI:10.1016/j.tim.2024.02.011 (Source: Trends in Microbiology)
