Prevalence of IRF4 rearrangement in large B-cell lymphomas of the Waldeyer ’s ring in adults
AbstractDiffuse large B-cell lymphoma (DLBCL) is the most common B-cell non-Hodgkin lymphoma (B-NHL) in adults. These lymphomas are classified according to gene expression profiling (GEP) into germinal center B-cell (GCB) and activated B-cell type (ABC). Recent studies have suggested new subtypes of large B-cell lymphoma, based on genetic and molecular alterations, among them is large B-cell lymphoma withIRF4-rearrangement (LBCL-IRF4). We used fluorescence in situ hybridization (FISH), GEP (using the DLBCL COO assay by HTG Molecular Inc), and next generation sequencing (NGS) to comprehensively characterize 30 cases of LBCL...

Subcutaneous chondromyxoid fibroma with a novel PNISR::GRM1 fusion —report of a primary soft tissue tumour without connection to an underlying bone
We report a subcutaneous CMF in a 34-year-old male, located on the distal medial aspect of the right thigh without any connection with the femur. The tumour measured 15  mm, it was well-circumscribed and displayed typical morphological features of a CMF. At the periphery, there was a small area of metaplastic bone. Immunohistochemically, the tumour cells were diffusely positive for smooth muscle actin and GRM1, and negative for S100 protein, desmin and cytokeratin AE1AE3. Whole transcriptome sequencing revealed a novelPNISR::GRM1 gene fusion. Our case indicates that CMF should be included in the differential diagnosis of s...

Discordance of PD-L1 expression in primary and metastatic ovarian high-grade serous carcinoma and its correlation with CD8  + tumor-infiltrating lymphocytes and patient prognosis
AbstractDifferential expression of programmed death-1 ligand (PD-L1) and its clinical significance in primary and metastatic ovarian high-grade serous carcinoma (HGSC) have not been defined. Thus, we investigated the PD-L1 expression of paired ovarian primary and omental metastatic HGSC and its correlation with CD8  + tumor-infiltrating lymphocyte (TILs) and patient survival. A total of 212 cases of ovarian HGSCs with matched primary ovarian and metastatic omental tumors accessioned between 2003 and 2018 were selected for further analysis. Using immunohistochemistry, we evaluated the density of CD8 + T ILs and expr...

Stromal grading predicts pathologic complete response and prognosis in triple-negative breast cancer
AbstractDo traditional prognostic factors fully account for the diversity of clinical behavior in breast cancer? Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer seen to have a poor prognosis, although there is great variation in clinical outcomes. Most recently, novel approaches have targeted the tumoral microenvironment (TME) to determine prognosis and tumor-associated stroma has become increasingly recognized as a potential biomarker to predict treatment response and prognosis in TNBC. The principle aim of this paper is to demonstrate an approach to stromal grading in TNBC, with considerati...
