Identification of miRNAs as non-invasive biomarkers for early diagnosis of lung cancers
AbstractCurrent clinical diagnostic methods lack the specificity in detecting lung cancer patients. The issue is particularly critical for stage I and II patients. Considerable evidence showed microRNA plays a very important role in lung carcinogenesis. Here, we identified a panel of 41 miRNAs significantly elevated in patients with lung cancer, of which eight miRNAs were further validated in an independent sample cohort. Classification analysis using the panel of eight miRNAs generated a discriminatory power of 93.3  % sensitivity and 93.8 % specificity in separating non-small-cell lung cancer (NSCLC) patients from norm...

MiR-212/132 is epigenetically downregulated by SOX4/EZH2-H3K27me3 feedback loop in ovarian cancer cells
In this study, we investigated the regulative effects of miR-212 and miR-132 on SOX4 expression in ovarian cancer cells and also studied whether there is a feedback regulation between miR-212/miR-132 and SOX4 via an epigenetic mechanism. The results showed that both EZH2 and SOX4 overexpressions significantly repressed miR-212 and miR-132 expressions in SKOV3 and OV2008 cells. Immunoprecipitation assay showed that there are interactions among SOX4, EZH2, and H3K27me3, and ChIP assay confirmed significant enrichment of EZH2 and H3K27me3 in the promoter region of miR-212/132. Both pri-miR-212 and pri-miR-132 expressions decr...

The E6-TAp63 β-Dicer feedback loop involves in miR-375 downregulation and epithelial-to-mesenchymal transition in HR-HPV+ cervical cancer cells
In this study, we found that miR-375 overexpression restored TAp63β expression. TAp63β overexpression significantly enhanced transcription and translation of Dicer, which furth er led to increased mature miR-375 levels. Therefore, we infer that there is an E6-TAp63β-Dicer feedback loop involved in miR-375 dysregulation in cervical cancer. Besides, we observed that enforced TAp63β expression significantly reduced the mesenchymal markers including N-cadherin, Vimentin, Sna il, and Slug but increased the epithelial marker E-cadherin in both Hela and SiHa cells. The wound healing assay also confirmed that TAp63β overexpre...

Establishment of a miRNA-mRNA regulatory network in metastatic renal cell carcinoma and screening of potential therapeutic targets
This study aimed to screen effective diagnosis or treatment biomarkers for renal cell carcinoma, especially for metastatic renal cell carcinoma (mRCC) based on microRNA (miRNA) and messenger RNA (mRNA) genechip, and their regulatory network. The differential expressions of miRNAs and mRNAs were examined by miRNA and mRNA gene-chip analyses, respectively, in patients with either localized renal cell carcinoma (lRCC) or mRCC, and a miRNA-mRNA regulatory network was established. Subsequently, the regulation of selected mRNAs by miRNAs was validated by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and ...
