Mapping morbidity 10 years prior to a diagnosis of young onset Alzheimer's disease
DISCUSSION: YOAD patients display a different pattern of morbidity up to 10 years prior to diagnosis. Awareness of specific alterations in morbidity may improve efforts toward a timely diagnosis.HIGHLIGHTS: Retrospective, nested case-control study of young onset Alzheimer's disease (YOAD). YOAD cases had a higher morbidity burden than controls. YOAD cases had a higher psychiatric morbidity burden up to 10 years before diagnosis. Altered morbidity patterns could serve as an early warning sign of YOAD.PMID:38294143 | DOI:10.1002/alz.13681 (Source: The Journal of Alzheimers Association)

Social frailty and the incidence of motoric cognitive risk syndrome in older adults
DISCUSSION: Social frailty was associated with an increased risk of incident MCR in older adults.HIGHLIGHTS: Various associations between social factors and motoric cognitive risk syndrome (MCR) have been reported. Social frailty that integrated from multiple social factors was associated with an increased risk of incident MCR. Social frailty should be included in the early screening of individuals to identify those at higher risk of MCR.PMID:38284799 | DOI:10.1002/alz.13696 (Source: The Journal of Alzheimers Association)

Cross-national comparisons of later-life cognitive function using data from the Harmonized Cognitive Assessment Protocol (HCAP): Considerations and recommended best practices
Alzheimers Dement. 2024 Jan 29. doi: 10.1002/alz.13694. Online ahead of print.ABSTRACTThe Harmonized Cognitive Assessment Protocol (HCAP) is a major innovation that provides, for the first time, harmonized data for cross-national comparisons of later-life cognitive functions that are sensitive to linguistic, cultural, and educational differences across countries. However, cognitive function does not lend itself to direct comparison across diverse populations without careful consideration of the best practices for such comparisons. This perspective discusses theoretical and methodological considerations and offers a set of ...

More than a piece of cake: Noun classifier processing in primary progressive aphasia
This study highlights noun classifiers as linguistic markers to discriminate PPA syndromes in Chinese speakers.HIGHLIGHTS: Noun classifier processing varies in the different primary progressive aphasia (PPA) variants. Specifically, semantic variant PPA (svPPA) and logopenic variant PPA (lvPPA) patients showed significantly lower ability in producing specific classifiers. Compared to lvPPA, svPPA patients were less able to choose the accurate classifiers when presented with choices. In svPPA, classifier production score was positively correlated with gray matter volume over bilateral temporal and left visual association cor...

Harmonizing florbetapir and PiB PET measurements of cortical A β plaque burden using multiple regions-of-interest and machine learning techniques: An alternative to the Centiloid approach
DISCUSSION: ML improved mcAβ comparability. Additional studies are needed for the generalizability to other amyloid tracers, and to tau PET. Highlights Centiloid is a calibration of the amyloid scale, not harmonization. Centiloid unifies the amyloid scale without improving inter-tracer association (R2 ). Machine learning (ML) can harmonize the amyloid scale by improving R2 . ML harmonization maps multi-regional florbetapir SUVRs to PiB mean-cortical SUVR. Artificial neural network ML increases Centiloid R2 from 86% to 97%.PMID:38276892 | DOI:10.1002/alz.13677 (Source: The Journal of Alzheimers Association)

C5aR1 signaling promotes region- and age-dependent synaptic pruning in models of Alzheimer's disease
DISCUSSION: Reduction of excessive synaptic pruning is an additional beneficial outcome of the suppression of C5a-C5aR1 signaling, further supporting its potential as an effective targeted therapy to treat AD.HIGHLIGHTS: C5aR1 ablation restores long-term potentiation in the Arctic model of AD. C5aR1 ablation rescues region specific excessive pre-synaptic loss. C5aR1 antagonist, PMX205, rescues VGlut1 loss in the Tg2576 model of AD. C1q tagging is not sufficient to induce VGlut1 microglial ingestion. Astrocytes contribute to excessive pre-synaptic loss at late stages of the disease.PMID:38278523 | DOI:10.1002/alz.13682 (Sou...

Down Syndrome Biobank Consortium: A perspective
Alzheimers Dement. 2024 Jan 25. doi: 10.1002/alz.13692. Online ahead of print.ABSTRACTIndividuals with Down syndrome (DS) have a partial or complete trisomy of chromosome 21, resulting in an increased risk for early-onset Alzheimer's disease (AD)-type dementia by early midlife. Despite ongoing clinical trials to treat late-onset AD, individuals with DS are often excluded. Furthermore, timely diagnosis or management is often not available. Of the genetic causes of AD, people with DS represent the largest cohort. Currently, there is a knowledge gap regarding the underlying neurobiological mechanisms of DS-related AD (DS-AD),...

Lewy body dementia: Overcoming barriers and identifying solutions
Alzheimers Dement. 2024 Jan 24. doi: 10.1002/alz.13674. Online ahead of print.ABSTRACTDespite its high prevalence among dementias, Lewy body dementia (LBD) remains poorly understood with a limited, albeit growing, evidence base. The public-health burden that LBD imposes is worsened by overlapping pathologies, which contribute to misdiagnosis, and lack of treatments. For this report, we gathered and analyzed public-domain information on advocacy, funding, research outputs, and the therapeutic pipeline to identify gaps in each of these key elements. To further understand the current gaps, we also conducted interviews with le...
