Aptamer inhibitor selection of SpyCas9 through CE-SELEX
Talanta. 2024 Mar 2;273:125837. doi: 10.1016/j.talanta.2024.125837. Online ahead of print.ABSTRACTCRISPR/Cas9 is a natural immune system of archaea and bacteria, which has been widely used in gene editing. In order to better control and improve the accuracy and safety of the system, inhibitors for SpyCas9 as "switches" have been selected for several years. The available inhibitors currently are all natural polypeptides inhibitors derived from phages, except one small molecule inhibitor. These natural inhibitors are challenging to obtain and are available in limited quantities, and the small molecule inhibitor is cytotoxic....

Fluorescent DNA-Silver nanoclusters in food safety detection: From synthesis to application
Talanta. 2024 Mar 8;273:125834. doi: 10.1016/j.talanta.2024.125834. Online ahead of print.ABSTRACTIn recent years, the conventional preparation of silver nanoclusters (AgNCs) has attracted much attention due to their ultra-small size, tunable fluorescence, easy-to-engineer, as well as biocompatible material. Moreover, its great affinity towards cytosine bases on single-stranded DNA has led to the construction of biosensors, especially aptamers, for a broad variety of applications in food safety and environmental protection. In past years, numerous researchers paid attention to the construction of AgNCs aptasensor. Therefor...

Online water vapor removal membrane inlet mass spectrometer for high-sensitivity detection of dissolved methane
In this study, we optimized the design of the sampling gas path and developed a high gas-tightness, high pressure-resistant membrane inlet system, coupled with a small-volume, low-power online water vapor removal system. This innovation efficiently eliminates water vapor while maintaining a high permeation flux of the target gases. By elevating the vacuum level to the order of 1E-6 Torr, the ionization efficiency and detection performance were improved. Based on this, we created an online water vapor removal membrane inlet mass spectrometer and conducted experimental research. Results indicated that the water removal effic...

Integrating of analytical techniques with enzyme-mimicking nanomaterials for the fabrication of microfluidic systems for biomedical analysis
Talanta. 2024 Mar 12;273:125896. doi: 10.1016/j.talanta.2024.125896. Online ahead of print.ABSTRACTBioanalysis faces challenges in achieving fast, reliable, and point-of-care (POC) determination methods for timely diagnosis and prognosis of diseases. POC devices often display lower sensitivity compared to laboratory-based methods, limiting their ability to quantify low concentrations of target analytes. To enhance sensitivity, the synthesis of new materials and improvement of the efficiency of the analytical strategies are necessary. Enzyme-mimicking materials have revolutionized the field of the fabrication of new high-th...

Tear metabolomics for the diagnosis of primary open-angle glaucoma
Talanta. 2024 Feb 25;273:125826. doi: 10.1016/j.talanta.2024.125826. Online ahead of print.ABSTRACTPrimary Open-Angle Glaucoma (POAG) is the most prevalent glaucoma type, and the leading cause of irreversible visual impairment and blindness worldwide. Identification of early POAG biomarkers is of enormous value, as there is not an effective treatment for the glaucomatous optic nerve degeneration (OND). In this pilot study, a metabolomic analysis, by using proton (1H) nuclear magnetic resonance (NMR) spectroscopy was conducted in tears, in order to determine the changes of specific metabolites in the initial glaucoma eyes a...

Sustainable beeswax modified cellulose paper for the determination of tricyclic antidepressants in biofluids
This article describes the synthesis of sorptive phases for bioanalysis based on the modification of cellulose paper with natural beeswax as sorbent, resulting in a substrate completely renewable and sustainable. The preparation of the sorptive phases consisted of the dissolution of beeswax in hexane, followed by its drop-casting on cellulose paper and subsequent evaporation of the solvent. The beeswax modification of paper renders it hydrophobic, enabling the extraction of the target analytes, i.e., imipramine, desipramine, amitriptyline and trimipramine, via hydrophobic interactions. The main variables affecting the extr...

Fast FRAP-SIA method to determine antioxidant capacity
Talanta. 2024 Feb 28;273:125813. doi: 10.1016/j.talanta.2024.125813. Online ahead of print.ABSTRACTOne of the most used methods to measure antioxidant capacity in food is the ferric reducing antioxidant power (FRAP) test, which is simple, sensitive, and economical, nevertheless has long analysis times, causing measurement errors due to the instability of the FRAP reagent due to its precipitation sequential injection analysis (SIA) is a flow technique that can correct these disadvantages because it is more quickly. So, a novel FRAP-SIA method was developed to evaluate the antioxidant capacity. The system was optimized using...

Development of double-layer poly (amino acid) modified electrochemical sensor for sensitive and direct detection of betamethasone in cosmetics
In this study, poly-L-Serine/poly-Taurine modified electrode (P(Tau)/P(L-Ser)/GCE) was prepared for sensitive and direct determination of betamethasone in cosmetics by a simple two-step in situ electropolymerization reaction. The relevant parameters of preparation and electroanalytical conditions were respectively studied, including the concentration of polymerization solution, the number of scanning circles and the scanning rate. The SEM and EDS mapping demonstrated successful preparation of P(Tau)/P(L-Ser)/GCE. The electro-catalytic properties of the obtained electrodes were investigated using cyclic voltammetry and diff...

A fluorescent probe for detecting H < sub > 2 < /sub > O < sub > 2 < /sub > and delivering H < sub > 2 < /sub > S in lysosomes and its application in maintaining the redox environments
Talanta. 2024 Mar 8;273:125894. doi: 10.1016/j.talanta.2024.125894. Online ahead of print.ABSTRACTHydrogen peroxide (H2O2) is a reactive oxygen species (ROS) that can be used as a marker for the occurrence of oxidative stress in the organism. Lysosomes serve as intracellular digestive sites, and when the concentration of H2O2 in them is abnormal, lysosomal function is often impaired, leading to the development of diseases. Hydrogen sulfide (H2S) acts as a gaseous signaling molecule that scavenges H2O2 from cells and tissues, thereby maintaining the redox environment of the body. However, most of the reported hydrogen perox...
