Exploring protein –protein interactions and oligomerization state of pulmonary surfactant protein C (SP‐C) through FRET and fluorescence self‐quenching
AbstractPulmonary surfactant (PS) is a lipid –protein complex that forms films reducing surface tension at the alveolar air–liquid interface. Surfactant protein C (SP-C) plays a key role in rearranging the lipids at the PS surface layers during breathing. The N-terminal segment of SP-C, a lipopeptide of 35 amino acids, contains two palmito ylated cysteines, which affect the stability and structure of the molecule. The C-terminal region comprises a transmembrane α-helix that contains a ALLMG motif, supposedly analogous to a well-studied dimerization motif in glycophorin A. Previous studies have demonstrated the potenti...

Molecular interactions between perlecan LG3 and the SARS ‐CoV‐2 spike protein receptor binding domain
In this study, we investigated the binding interactions between the SARS-CoV-2 spike protein RBD and perlecan LG3 through molecular modeling simulations and surface plasmon resonance (SPR) experiments. Our results indicate stable binding between LG3 and SARS-CoV-2 spike protein RBD, which may potentially enhance RBD-ACE2 interactions. These findings shed light on the role of perlecan LG3 in SARS-CoV-2 infection and provide insight into SARS-CoV-2 pathophysiology and potential therapeutic strategy for COVID-19. TEPCAM: Prediction of T ‐cell receptor–epitope binding specificity via interpretable deep learning
AbstractThe recognition of T-cell receptor (TCR) on the surface of T cell to specific epitope presented by the major histocompatibility complex is the key to trigger the immune response. Identifying the binding rules of TCR –epitope pair is crucial for developing immunotherapies, including neoantigen vaccine and drugs. Accurate prediction of TCR–epitope binding specificity via deep learning remains challenging, especially in test cases which are unseen in the training set. Here, we propose TEPCAM (TCR–EPitope ide ntification based on Cross-Attention and Multi-channel convolution), a deep learning model that incorpora...
