Mechanisms impairing blood pressure responses to nitrite and nitrate
Publication date: Available online 1 February 2019Source: Nitric OxideAuthor(s): Gustavo H. Oliveira-Paula, Lucas C. Pinheiro, Jose E. Tanus-SantosAbstractHypertension is a multifactorial disease associated with impaired nitric oxide (NO) production and bioavailability. In this respect, restoring NO activity by using nitrite and nitrate has been considered a potential therapeutic strategy to treat hypertension. This possibility is justified by the understanding that both nitrite and nitrate may be recycled back to NO and also promote the generation of other bioactive species. This process involves a complex biological circ...

Acceleration of the autoxidation of nitric oxide by proteins
Publication date: Available online 30 January 2019Source: Nitric OxideAuthor(s): Matías N. Möller, Ana DenicolaAbstractLipoproteins and lipid membranes accelerate •NO autoxidation by increasing local concentration of •NO and O2. Although the idea that proteins could also accelerate this reaction was presented some time ago, it was largely criticized and dismissed. Herein the effect of proteins on •NO autoxidation rates was studied following •NO disappearance with a selective electrode. It was found that human serum albumin (HSA) accelerated •NO autoxidation by a factor of 9 per g/mL of protein, much less than p...

The era of nitric oxide in plant biology: Twenty years tying up loose ends
Publication date: Available online 28 January 2019Source: Nitric OxideAuthor(s): Fiorella Del Castello, Andrés Nejamkin, Raul Cassia, Natalia Correa-Aragunde, Belén Fernández, Noelia Foresi, Cristina Lombardo, Leonor Ramirez, Lorenzo LamattinaAbstractNitric oxide (NO) is an essential signal molecule to maintain cellular homeostasis in uni and pluricellular organisms. Conceptually, NO intervenes as much in sustaining basal metabolic processes, as in firing cellular responses to changes in internal and external conditions, and also in guiding the return to basal conditions. Behind these unusual capabilities of NO is the c...

Chronic high-dose beetroot juice supplementation improves time trial performance of well-trained cyclists in normoxia and hypoxia
Publication date: Available online 24 January 2019Source: Nitric OxideAuthor(s): Torben Rokkedal-Lausch, Jesper Franch, Mathias K. Poulsen, Lars P. Thomsen, Eddie Weitzberg, Ernest N. Kamavuako, Dan S. Karbing, Ryan G. LarsenAbstractDietary nitrate (NO3−) supplementation via beetroot juice (BR) is known to improve endurance performance in untrained and moderately trained individuals. However, conflicting results exist in well-trained individuals. Evidence suggests that the effects of NO3− are augmented during conditions of reduced oxygen availability (e.g., hypoxia), thereby increasing the probability of performance im...

Effects of acute dietary nitrate supplementation on aortic blood pressures and pulse wave characteristics in post-menopausal women
ConclusionsDietary nitrate supplementation favorably modifies aortic systolic and mean blood pressure under resting conditions in healthy postmenopausal women. Acute supplementation of nitrate does not, however, appear to restore indices of aortic stiffness in this group. Future work should evaluate chronic, long-term effects of this non-pharmacological supplement. (Source: Nitric Oxide)

Dexmedetomidine protects against lipopolysaccharide-induced early acute kidney injury by inhibiting the iNOS/NO signaling pathway in rats
Publication date: Available online 17 January 2019Source: Nitric OxideAuthor(s): Yongping Chen, Li Luan, Chaoran Wang, Manyu Song, Yuan Zhao, Yujie Yao, Haotian Yang, Biao Ma, Honggang FanAbstractIncreasing evidence has demonstrated that dexmedetomidine (DEX) possesses multiple pharmacological actions. Herein, we explored the protective effect and potential molecular mechanism of DEX on lipopolysaccharide (LPS)-induced early acute kidney injury (AKI) from the perspective of antioxidant stress. We found that DEX (30 μg/kg, i.p.) ameliorated the renal dysfunction and histopathological damage (tubular necrosis, vacuolar de...

Encapsulation of S-nitrosoglutathione into chitosan nanoparticles improves drought tolerance of sugarcane plants
Publication date: 1 March 2019Source: Nitric Oxide, Volume 84Author(s): Neidiquele M. Silveira, Amedea B. Seabra, Fernanda C.C. Marcos, Milena T. Pelegrino, Eduardo C. Machado, Rafael V. RibeiroAbstractThe entrapment of NO donors in nanomaterials has emerged as a strategy to protect these molecules from rapid degradation, allowing a more controlled release of NO and prolonging its effect. On the other hand, we have found beneficial effects of S-nitrosoglutathione (GSNO) – a NO donor – supplying to sugarcane plants under water deficit. Here, we hypothesized that GSNO encapsulated into nanoparticles would be more effecti...

Pharmacokinetics and pharmacodynamics of nitric oxide mimetic agents
Publication date: Available online 11 January 2019Source: Nitric OxideAuthor(s): Austin Horton, Isaac T. SchieferAbstractDrug discovery focusing on NO mimetics has been hamstrung due to its unconventional nature. Central to these challenges is the fact that direct measurement of molecular NO in biological systems is exceedingly difficulty. Hence, drug development of NO mimetics must rely upon measurement of the NO donating specie (i.e., a prodrug) and a downstream marker of efficacy without directly measuring the molecule, NO, that is responsible for biological effect. The focus of this review is to catalog in vivo attempt...

Association of endothelial nitric oxide synthase gene G894T polymorphism with blood oxygen transport
ConclusionThe T-allele of G894T polymorphism is associated with low values for oxygen content, oxygen tension, acidic pH shift and increased haemoglobin affinity for oxygen under standard and real conditions. The presence of a minor allele in G894T polymorphism of the NOS3 gene contributes to formation of oxygen transport function of blood. (Source: Nitric Oxide)
