Trajectories of CSF and plasma biomarkers across Alzheimer's disease continuum: disease staging by NF-L, p-tau181, and GFAP
Neurobiol Dis. 2023 Nov 15;189:106356. doi: 10.1016/j.nbd.2023.106356. Online ahead of print.ABSTRACTCSF-to-plasma transition will open new avenues for molecular phenotyping of Alzheimer's disease (AD). Here we evaluated a panel of AD biomarkers in matched CSF and plasma samples across the AD continuum, from preclinical AD to dementia. The aims were to: 1) compare diagnostic performance of the two biofluids, 2) evaluate trajectories of the biomarkers along AD progression. We analyzed CSF and plasma Aβ42/40, p-tau181, p-tau231, t-tau, NF-L, GFAP, UCHL-1 and CSF SNAP-25 in a cohort (n = 173) of preclinical AD, MCI-AD, AD de...

Polygenic risk scores for Alzheimer's disease in relation to cognitive change: A representative sample from the general population followed over 16  years
CONCLUSION: We found that genetic predisposition of AD predicted cognitive decline among 70-year-olds followed over 16 years, regardless of dementia status, while polygenic risk for general cognitive performance did not.PMID:37977433 | DOI:10.1016/j.nbd.2023.106357 (Source: Neurobiology of Disease)

Projection-defined median raphe Pet < sup > + < /sup > subpopulations are diversely implicated in seizure
Neurobiol Dis. 2023 Nov 15;189:106358. doi: 10.1016/j.nbd.2023.106358. Online ahead of print.ABSTRACTThe raphe nuclei, the primary resource of forebrain 5-HT, play an important but heterogeneous role in regulating subcortical excitabilities. Fundamental circuit organizations of different median raphe (MR) subsystems are far from completely understood. In the present study, using cell-specific viral tracing, Ca2+ fiber photometry and epilepsy model, we map out the forebrain efferent and afferent of different MR Pet+ subpopulations and their divergent roles in epilepsy. We found that PetMR neurons send both collateral and pa...

Hippocampal seizures differentially modulate locus coeruleus activity and result in consistent time-locked release of noradrenaline in rat hippocampus
In conclusion, this study showed consistent but heterogeneous modulation of LC neurons and a consistent time-locked release of hippocampal noradrenaline during acute hippocampal seizures.PMID:37977430 | DOI:10.1016/j.nbd.2023.106355 (Source: Neurobiology of Disease)

Gray matter structural covariance networks patterns associated with autopsy-confirmed LATE-NC compared to Alzheimer's disease pathology
CONCLUSIONS: This study showed different SCN patterns in the cases with LATE-NC, AD, and AD+LATE-NC, and indicated the network disconnection mechanism underlying these three neuropathological progressions. Further, SCN may serve as an effective biomarker to distinguish between different types of dementia.PMID:37977431 | DOI:10.1016/j.nbd.2023.106354 (Source: Neurobiology of Disease)
