Etoposide-induced DNA damage in a chromosomal breakpoint of RUNX1 gene is independent of RUNX1 expression
Publication date: Available online 27 August 2019Source: Leukemia Research ReportsAuthor(s): Nicolás Schnake, Soraya GutiérrezAbstractIn this work, we analyzed the association between RUNX1 gene expression and the accessibility of BCR3, one of RUNX1 gene breakpoint regions involved in the chromosomal translocation (8;21), a frequent translocation in treatment-related acute myeloid leukemia patients. To this end, we evaluate DNA damage generation induced by in vitro etoposide treatment of KG-1 and Colo320 cells. Our results show that treatment using clinical doses of etoposide for 24 hours induces the generation of DNA do...

Prognostic Significance of Fc Receptor-like 1 in Patients with Chronic Lymphocytic Leukemia, Hairy Cell Leukemia, and Various B-cell Non-Hodgkin's Lymphoma
This study examined the expression pattern of FCRL1 in B-cell non-Hodgkin's lymphoma patients using real-time PCR and flow cytometry. The results revealed higher levels of FCRL1 expression in diffuse large B-cell lymphoma, hairy cell leukemia, and Burkitt lymphoma patients compared with control groups. There was a significant reduction in the levels of FCRL1 expression in chronic lymphocytic leukemia and mantle cell lymphoma patients compared with healthy individuals. These findings suggest FCRL1 as an excellent marker for the prognosis or immunotherapy of B-cell malignancies. (Source: Leukemia Research Reports)

TTP-like syndrome associated with Hemoglobin SC disease treated successfully with plasma and red cell exchange
ConclusionAlthough literature showed therapeutic plasma exchange alone can abrogate a similar clinical scenario, we did an in-depth review which concluded that in most of the TMA cases secondary to sickle cell disease, treatment with both with plasma exchange and red cell exchange transfusion are necessary. Rapid Progression to AML in a Patient with Germline GATA2 Mutation and Acquired NRAS Q61K Mutation
We present a patient with GATA2 mutation and MDS who progressed to AML over four months. Whole exome and targeted deep sequencing identified a new p.Q61K NRAS mutation in the bone marrow at the time of AML development. Rapid development of AML is possible in the setting of germline GATA2 mutation despite stable MDS, supporting close monitoring and consideration of early allogeneic transplantation. A rare case of Acute Myeloid Leukemia with der(1)t(1;19)(p13;p13.1)
We present a case of acute myeloid leukemia with der(1)t(1;19)(p13;p13.1) translocation and RUNX1 mutation. A literature review summarizing the clinical, pathological, and molecular features of the published cases is also presented. (Source: Leukemia Research Reports)

Intracranial acute promyelocytic leukemia at presentation—A case-based discussion
Publication date: 2019Source: Leukemia Research Reports, Volume 11Author(s): Kristin Sticco, Tahmeena Ahmed, Huda SalmanAbstractIntracranial disease is a very rare presentation at diagnosis in acute promyelocytic leukemia (APL). The risk associated with this particular presentation is not accounted for when the current risk stratification is based on peripheral counts. Extra medullary disease in general may challenge this risk stratification that commands initial induction treatment of this potentially fatal disease. Here we discuss a case presented at diagnosis with extensive intracranial base of the skull, clivus and sin...

Neutrophil-to-lymphocyte ratio (NLR) fails to predict outcome of diffuse large B cell lymphoma
Conclusion: Our study failed to reveal the predictive value of NLR and demonstrated that the NCCN-IPI might be the most powerful predictor in DLBCL. (Source: Leukemia Research Reports)

Atypical Chronic Myeloid Leukemia BCR-ABL 1 negative: A case report and literature review
Publication date: Available online 25 May 2019Source: Leukemia Research ReportsAuthor(s): Jihane Belkhair, Abderahim Raissi, Hicham Elyahyaoui, Mustapha Ait Ameur, Mohamed ChakourAbstractAtypical chronic myeloid leukemia (aCML), BCR-ABL1 negative is a rare myelodysplastic syndrome /myeloproliferative neoplasm for which no current standard of care exists. The blood smear of patients with aCML showed prominent immature granulocytosis, and granulocytic dysplasia. We admitted a 58-year-old man with splenomegaly, hyperleukocytosis, anemia, and thrombocytopenia; then cytology, cytogenetic and molecular biology analysis of bone m...

Tyrosine kinase inhibitor-induced carotid stenosis: a case report
Publication date: Available online 25 May 2019Source: Leukemia Research ReportsAuthor(s): Jeanne Hersant, Martine Gardembas, Georges Leftheriotis, Patrick Vandeputte, Pierre Abraham, Samir HenniAbstractTyrosine kinase inhibitors are considered as highly effective and relatively safe drugs for the treatment of chronic myeloid leukemia. If several side effects on short and long term are well known and described, their involvement in the development of carotid stenosis remains unclear. Here, we describe a case of carotid stenosis in a patient receiving tyrosine kinase inhibitors and discuss the current literature. (Source: Le...

A Rare Case Of Near-Haploid Acute Lymphoblastic Leukemia
We report a case in an 18-year old male. He presented with two abnormal clones, one with 27∼28 and one with 54∼56 chromosomes. Near-haploidy (27∼28) carries a poor prognosis and hyperdiploidy (>50) has a good prognosis. The correct diagnosis was critical for this patient's prognosis and treatment. The patient achieved remission after a bone marrow transplant from a 10/10 HLA matched sibling donor. He relapsed six months later and expired seven months later. This case illustrates the need for careful standard and molecular cytogenetic analysis for accurate diagnosis and treatment for this rare type of ALL. (Source: Le...
