Biomimetic fetal rotation bioreactor for engineering bone tissues —Effect of cyclic strains on upregulation of osteogenic gene expression
This study introduces a multimodal bioreactor system that allows application of cyclic compressive strains on premature bone grafts that are cultured under biaxial rotation (chamber rotation about 2 axes) conditions for bone tissue engineering. The bioreactor is integrated with sensors for dissolved oxygen levels and pH that allow real‐time, non‐invasive monitoring of the culture parameters. Mesenchymal stem cells‐seeded polycaprolactone–β‐tricalcium phosphate scaffolds were cultured in this bioreactor over 2 weeks in 4 different modes—static, cyclic compression, biaxial rotation, and multimodal (combination ...

Decellularized human liver extracellular matrix (hDLM) ‐mediated hepatic differentiation of human induced pluripotent stem cells (hIPSCs)
Abstract Liver tissue engineering has emerged as a promising approach in organ transplantation but has been hampered by the lack of a reliable and readily available cell source. Human induced pluripotent stem cells hiPSCs have been highlighted as a desirable source, due to their differentiation potential, ability to self‐renew, and the possibility of making patient‐specific cells. We developed a decellularization protocol that efficiently removes cellular material while retaining extracellular matrix components. Subsequently, hiPSCs were differentiated on decellularized human liver extracellular matrix (hDLM) scaffolds...

Regenerative effects of human embryonic stem cell ‐derived neural crest cells for treatment of peripheral nerve injury.
In this study, neural crest cells were differentiated from human embryonic stem cells. The differentiated cells exhibited typical stellate morphology and protein expression signatures that were comparable to native neural crest. Conditioned media harvested from the differentiated cells contained a range of biologically active trophic factors and was able to stimulate in vitro neurite outgrowth. Differentiated neural crest cells were seeded into a biodegradable nerve conduit and their regeneration potential was assessed in a rat sciatic nerve injury model. A robust regeneration front was observed across the entire width of ...

An endochondral ossification approach to early ‐stage bone repair: Use of tissue‐engineered hypertrophic cartilage constructs as primordial templates for weight‐bearing bone repair
In this study, we demonstrate that tissue‐engineered hypertrophically‐primed cartilage constructs can be developed from collagen‐based scaffolds cultured with mesenchymal stem cells. These constructs were subsequently implanted into femoral defects in rats. It was evident that the constructs could support enhanced early‐stage healing at 4 weeks of these weight‐bearing femoral bone defects compared to untreated defects.
