Perivascular invasion of primary human glioblastoma cells in organotypic human brain slices: human cells migrating in human brain
ConclusionHuman organotypic models can accurately study cell migration  ex vivo. GBM IDH-wild type cells migrate toward the perivascular space in blood vessels and their migratory parameters change once they contact vascular structures and under hypoxic conditions. This model allows the evaluation of GBM invasion, considering the human brain microenvironment when cell s are removed from their native niche after surgery.Graphical Abstract (Source: Journal of Neuro-Oncology)

Tucatinib and stereotactic radiosurgery in the management of HER2 positive breast cancer brain metastases
ConclusionsSRS in combination with tucatinib, capecitabine, and trastuzumab appears to be a safe and feasible treatment for HER2  + brain metastases. Further prospective evaluation of potential synergistic effects is warranted. (Source: Journal of Neuro-Oncology)

Effect of race, sex, and socioeconomic factors on overall survival following the resection of intramedullary spinal cord tumors
ConclusionOur study sheds light on the healthcare disparities that exist in the surgical management of IMSCTs. Our findings indicate that race, sex, socioeconomic status, and treatment facility are independent predictors of 5  year mortality, with Black patients, males, those with lower socioeconomic status, and those treated at non-academic centers experiencing significantly higher mortality rates. These alarming disparities underscore the urgent need for policymakers and researchers to address the underlying factors c ontributing to these discrepancies and provide equal access to high-quality surgical care for patients ...

Molecular predictors for decitabine efficacy in meningiomas – a pilot study
ConclusionEarly effects of DCT in meningiomas are strongly related with DNMT1 expression, while clinical, histological, and molecular predictors for efficacy are sparse. Kinetics of drug efficacy might indicate necessity of repeated exposition and encourage further analyses. (Source: Journal of Neuro-Oncology)

Efficacy of intrathecal methotrexate in children with high-risk medulloblastoma over three years: a retrospective study from a single center
ConclusionsOur findings support the addition of intrathecal MTX during chemotherapy as the optimal management for children with group 3 and SHH high-risk MB. However, it is not recommended for group 4  MB patients, especially in resource-limited regions.Trial registration number: Retrospective registered No.(2020  − 117). Stereotactic radiosurgery for patients with brain metastases from gastroesophageal cancers
ConclusionsIn this study, the 12-month local tumor control rate was 90%. Incidences of adverse radiation effect rates were rare. The median overall survival of 7 months indicates the poor prognosis of patients with brain spread of their GE cancer. (Source: Journal of Neuro-Oncology)

Significance of O6-methyl guanine methyltransferase promoter methylation in high grade glioma patients: optimal cutoff point, CpG locus, and genetic assay
ConclusionOur outcomes support the prognostic value ofMGMT promoter methylation in patients with high grade glioma. Sequencing of whole promoter CpG islands demonstrated that methylation of particular CpG sites might predict clinical outcomes more precisely. (Source: Journal of Neuro-Oncology)

Cellular senescence in glioma
ConclusionIn this article, we delineate an overview of senescence in the context of gliomas, including the mechanisms that lead to senescence induction, bypass, and escape. Furthermore, we examine the role of senescent cells in the tumor microenvironment and their role in tumor progression and recurrence. Additionally, we highlight potential therapeutic opportunities for targeting senescence in glioma. (Source: Journal of Neuro-Oncology)

Hypo-fractionated accelerated radiotherapy with concurrent and maintenance temozolomide in newly diagnosed glioblastoma: updated results from phase II HART-GBM trial
ConclusionThe long-term results of the trial support HART as a promising treatment option with comparable survival outcomes to the current standard of care. Phase III trials are required for further validation of this regimen which has the potential to become the new standard of care in GBM. (Source: Journal of Neuro-Oncology)

Belzutifan in adults with VHL-associated central nervous system hemangioblastoma: a single-center experience
ConclusionWe showed that belzutifan is safe and well-tolerated with strong disease response for CNS hemangioblastoma in adults with VHL, supporting continued use of belzutifan in this patient population. Future studies should assess duration of treatment, effects of cessation after long-term use, and markers of therapeutic response. (Source: Journal of Neuro-Oncology)
