-HH and -HAAAH motifs act as fishing nets for biologically relevant metal ions in metallopeptides
J Inorg Biochem. 2023 Dec 19;252:112456. doi: 10.1016/j.jinorgbio.2023.112456. Online ahead of print.ABSTRACTHistidine are one of the most common residues involved in transition metal ion binding in the active sites of metalloenzymes. In order to mimic enzymatic metal binding sites, it is crucial to understand the basic coordination modes of histidine residues, distributed at different positions in the peptide sequence. We show that: (i) the separation of two histidines has a large effect on complex stability - a sequence with adjusting histidine residues forms more stable complexes with Zn(II) than the one in which the re...

Enhancing ferryl accumulation in H < sub > 2 < /sub > O < sub > 2 < /sub > -dependent cytochrome P450s
We report the characterization of a highly chemoselective CYP decarboxylase from Staphylococcus aureus (OleTSA) that is soluble at high concentrations. Examination of OleTSA Compound I (CpdI) accumulation with a variety of fatty acid substrates reveals a dependence on resting spin-state equilibrium. Alteration of this equilibrium through targeted mutagenesis of the proximal pocket favors the high-spin form, and as a result, enhances Cpd-I accumulation to nearly stoichiometric yields.PMID:38141432 | DOI:10.1016/j.jinorgbio.2023.112458 (Source: Journal of Inorganic Biochemistry)

Effects of removal of the axial methionine heme ligand on the binding of S. cerevisiae iso-1 cytochrome c to cardiolipin
J Inorg Biochem. 2023 Dec 18;252:112455. doi: 10.1016/j.jinorgbio.2023.112455. Online ahead of print.ABSTRACTThe cleavage of the axial S(Met) - Fe bond in cytochrome c (cytc) upon binding to cardiolipin (CL), a glycerophospholipid of the inner mitochondrial membrane, is one of the key molecular changes that impart cytc with (lipo)peroxidase activity essential to its pro-apoptotic function. In this work, UV - VIS, CD, MCD and fluorescence spectroscopies were used to address the role of the Fe - M80 bond in controlling the cytc-CL interaction, by studying the binding of the Met80Ala (M80A) variant of S. cerevisiae iso-1 cytc...

A voltammetric peptide biosensor for Cu < sup > 2+ < /sup > metal ion quantification in coffee seeds
J Inorg Biochem. 2024 Feb;251:112441. doi: 10.1016/j.jinorgbio.2023.112441. Epub 2023 Nov 29.ABSTRACTA prion-derived copper(II)-binding peptide was assembled onto a gold electrode for the building of a voltammetric biosensor for measuring the Cu2+ metal ion in biological samples. The chosen sequence was H-CVNITKQHTVTTTT-NH2, with an appended cysteine residue for binding to the gold surface as a self-assembled monolayer and a histidine residue as the anchorage point for copper(II) complexation. The biosensor showed a linear range of 10-7 to 10-6 M with an 8.0 × 10-8 M detection limit and a 1.0 × 10-7 M quantification limi...
