The Synergism of Public Health Policies and Social Work May Reduce Mortality from Cancer, Liver Disease and Cardiovascular Disease
This study not only sheds light on the critical role of stringent alcohol-related public health policies in reducing mortality rates associated with alcohol-related disorders but also emphasize the potential of such policies to significantly impact the social determinants of health. Autoimmune Hepatitis: Mycophenolate mofetil versus azathioprine as first line therapy
A recent open label, multicenter, randomized, controlled trial by Snijders et al.1 compared the efficacy and safety of mycophenolate mofetil (MMF) and azathioprine (AZA) as first-line induction therapy in treatment naive autoimmune hepatitis (AIH). The main findings were that MMF showed better tolerability and higher rate of biochemical response at 24 weeks. The study is a welcome addition to the limited number of randomized controlled trials in AIH. The authors present clear data but we think that several issues need further clarification. Rare monogenic causes of steatotic liver disease masquerading as MASLD
With interest we read the Delphi consensus statement on the new fatty liver disease nomenclature.1 The strong link between non-alcoholic fatty liver disease and metabolic dysfunction/insulin resistance has resulted in the inclusion of cardiometabolic risk factors as diagnostic criteria of metabolic dysfunction associated steatotic liver disease (MASLD). When these risk factors are absent, other specific aetiologies of steatotic liver disease, such as monogenic diseases, should be considered.1 (Source: Journal of Hepatology)

Familial Coaggregation of MASLD and Liver-related Outcomes
Dr. Ebrahimi has served as an advisory board member for Boehringer Ingelheim. Dr Ludvigsson has coordinated an unrelated study on behalf of the Swedish IBD quality register (SWIBREG). That study received funding from Janssen Corporation. Dr Ludvigsson has also received financial support from MSD to develop a paper reviewing national healthcare registers in China and is currently discussing potential research collaboration on celiac disease with Takeda. Multi-omics characterization of healthy and PSC human liver – what we knew and what we have learned
To date, we have not fully understood the pathogenesis of primary sclerosing cholangitis (PSC), a dreadful progressive cholestatic disease associated with an increased risk of bacterial cholangitis and malignancies, such as cholangiocarcinoma and colorectal carcinoma, for which effective treatment options are lacking.1,2 Several reports and genetic association studies have pointed toward a dysregulation of the adaptive immune response in PSC,3 –5 but deeper functional insights into the immune pathogenesis have been hampered by the availability of sufficient amounts of human liver tissue, especially from early-disease sta...

PTEN Deficiency Induces an Extrahepatic Cholangitis-Cholangiocarcinoma Continuum via Aurora kinase A in Mice
The PTEN-AKT pathway is frequently altered in extrahepatic cholangiocarcinoma (eCCA). We aim to evaluate the role of PTEN in the pathogenesis of eCCA and find novel therapies for this disease. (Source: Journal of Hepatology)

Using liver stiffness to predict and monitor the risk of decompensation and mortality in patients with alcohol-related liver disease
Liver stiffness measurements (LSM) are recommended for disease prognostication and monitoring. We evaluated if LSM, using transient elastography, and LSM changes predict decompensation and mortality in patients with alcohol-related liver disease (ALD). (Source: Journal of Hepatology)

Single cell landscape of functionally cured chronic hepatitis B patients reveals activation of innate and altered CD4-CTL-driven adaptive immunity
In this study we comprehensively interrogate disease-associated cell states (DACS) identified within intra-hepatic tissue and matched PBMCs from either CHB or FC patients, at the resolution of single cells, to provide novel insights into putative mechanisms underlying FC. The Transition from NAFLD to MASLD and Its Impact on Clinical Practice and Outcomes
We read the study by Younossi et al1 with great interest and would like to congratulate the authors for their superb study. Over the last decade, due to changes in lifestyle, there's been a worldwide uptick in metabolic disorders such as obesity and type 2 diabetes, which have directly or indirectly impacted around 37% of the global adult population with non-alcoholic fatty liver disease (NAFLD).2 This condition is also recognized as a leading cause of hepatocellular carcinoma (HCC).3 However, given the insufficient understanding of NAFLD and the potential misconceptions its name may cause, a Delphi conference conducted by...

Autoimmune hepatitis: Azathioprine or mycophenolate mofetil for inducing remission?
We read with interest the first randomized-controlled trial evaluating the safety and efficacy of mycophenolate mofetil (MMF) as the first-line over azathioprine (AZA) for induction of adult-onset autoimmune hepatitis (AIH)1 and would like to raise the following points.
