In order to better understand hepatitis delta virus (HDV) prevalence at the population level in 25 countries/territories, The Polaris Observatory Collaborators conducted a literature review for anti-HDV and HDV RNA-positive prevalence in HBsAg-positive individuals and held virtual meetings with experts to discuss the findings and collect unpublished data. The findings were then combined with The Polaris Observatory HBV data to estimate the anti-HDV and HDV RNA prevalence in each country/territory at the population level. Reply to: “Outcome of in situ split liver transplantation in Italy over the last 25 years: An alternative analysis and personal view”
We thank Jean de Ville de Goyet1 for his comment on our study recently published in the Journal of Hepatology.2 Our study on outcomes of in situ split liver transplantation (SLT) in Italy confirms, on a larger population, the hypothesis proposed by de Ville de Goyet himself some time ago based on 100 SLTs from nine European centres,3 i.e. that improved donor-to-recipient (D/R) matching plays a key role in outcome improvement, a conclusion of which we are still firmly convinced. Merits and boundaries of the BCLC staging and treatment algorithm: Learning from the past to improve the future with a novel proposal
In this Expert Opinion, we thoroughly analyse the Barcelona Clinic Liver Cancer (BCLC) staging and treatment algorithm for hepatocellular carcinoma (HCC) that, since 1999, has standardised HCC management, offering a structured approach for the prognostic evaluation and treatment of patients with HCC. The first part of the article presents the strengths and evolutionary improvements of the BCLC staging system. Nevertheless, both patient characteristics and available treatments have changed in the last two decades, limiting the role of the BCLC criteria for treatment allocation in a growing number of patients. This Expert Opinion article thoroughly analyses the Barcelona Clinic Liver Cancer (BCLC) staging and treatment algorithm for hepatocellular carcinoma (HCC) that, since 1999, has standardised the HCC management, offering a structured approach for the prognostic evaluation and treatment recommendations of patients with HCC. The first part of the article presents the strengths and evolutionary improvements of the BCLC staging system. Molecular force-induced liberation of transforming growth factor-beta remodels the spleen for ectopic liver regeneration
Ectopic liver regeneration in the spleen is a promising alternative to organ transplantation for treating liver failure. To accommodate transplanted liver cells, the splenic tissue must undergo structural changes to increase extracellular matrix (ECM) content, demanding a safe, efficient approach for tissue remodelling. A gold mine of information from a deep dive into the liver transcriptome
Understanding the changes in hepatic metabolic pathways in response to drugs and dietary changes has been an important long-term goal. A comprehensive functional understanding requires identification of the proteomic and enzymatic changes in hundreds of biochemical reactions which is a daunting and currently technically unattainable task. However, quantification of changes in the RNA pool is much more practical, and due to the strict hierarchical organization of DNA being a template for RNA, and RNA being a template for protein; changes in RNA may correlate with protein levels and function. Optimal hepatitis C treatment adherence patterns and sustained virologic response among people who inject drugs: The HERO study
Direct-acting antivirals (DAAs) are highly effective for treating HCV infection even among people who inject drugs (PWID). Yet, little is known about patients' adherence patterns and their association with sustained virologic response (SVR) rates. We aimed to summarize various adherence patterns and determine their associations with SVR. (Source: Journal of Hepatology)

Radiotherapy following Intrahepatic Progression on Immunotherapy in Advanced Hepatocellular Carcinoma
We read with great interest of the recently published article “Combination immunotherapy for hepatocellular carcinoma” by Rimassa et al. which comprehensively reviewed the latest evidence of immunotherapy in advanced hepatocellular carcinoma (HCC).1 The authors provided important insights that, as now immunotherapy has moved to the first-line setting, the optimal management following immunotherapy progression is currently not defined. Organoid-guided Precision Hepatology for Metabolic Liver Disease
Metabolic dysfunction associated steatotic liver disease affects millions of people worldwide. Progress towards a definitive cure has been incremental and limited to only lifestyle modification. Hepatocyte-specific lipid accumulation is the main trigger of lipotoxic events, driving inflammafibrotic processes. The underlying pathology is extraordinarily heterogenous, and steatohepatitis manifestations are markedly influenced by metabolic communications across non-hepatic organs. Synthetic human tissue models have emerged as powerful platforms to better capture the mechanistic diversity in disease progression, while preservi...

A Novel Treatment Strategy Utilizing Panobinostat for High-Risk and Treatment-Refractory Hepatoblastoma
Patients with metastatic, treatment-refractory, and relapsed hepatoblastoma (HB) have survival rates of less than 50% due to limited treatment options. To develop new therapeutic strategies for these patients, our lab has developed a preclinical testing pipeline. Given that histone deacetylase (HDAC) inhibition has been proposed for HB, we hypothesized that we could find an effective combination treatment strategy utilizing HDAC inhibition.
