Effect of the deubiquitination enzyme gene UBP6 on the stress-responsive transcription factor Msn2-mediated control of the amino acid permease Gnp1 in yeast
Publication date: Available online 19 October 2019Source: Journal of Bioscience and BioengineeringAuthor(s): Noreen Suliani binti Mat Nanyan, Daisuke Watanabe, Yukiko Sugimoto, Hiroshi TakagiIn the yeast Saccharomyces cerevisiae, the transcriptional factor Msn2 plays an essential role in response to a variety of environmental stresses by activating the transcription of many genes that contain the stress-responsive elements in the promoters. We previously reported that overexpression of the MSN2 gene confers tolerance to various stresses in industrial yeast strains. Recently, the overexpression of MSN2 was shown to increase...

Simultaneous production of cellulase and ferulic acid esterase by Penicillium decumbens with rice straw as the sole carbon source
In this study, it was confirmed that Penicillium decumbens produces more ferulic acid esterase (FAE) than other cellulase-producing fungi. Cellulose, destarched wheat bran (DSWB), and rice straw were used as carbon sources. Little phenolic acid was released by cellulose- and DSWB-based enzymes during the saccharification of rice straw, whereas rice straw was a favorable carbon source for the simultaneous production of cellulase and FAE. High-performance liquid chromatography showed that during enzyme production, phenolic acids were released from rice straw, and ball-milling affected this release of phenolic acids. Small am...

Novel anti-obesity peptide (RLLPH) derived from hazelnut (Corylus heterophylla Fisch) protein hydrolysates inhibits adipogenesis in 3T3-L1 adipocytes by regulating adipogenic transcription factors and adenosine monophosphate-activated protein kinase (AMPK) activation
In this study, we purified and identified anti-obesity peptides from hazelnut protein hydrolysates by chromatography and liquid chromatography-tandem mass spectrometry (LC-MS/MS). Subsequently, we evaluated the inhibitory effect of the synthetic peptide on adipogenesis in 3T3-L1 adipocytes by Oil Red O staining, reverse transcription polymerase chain reaction (RT-PCR) and western blot. The results showed that a novel synthetic pentapeptide, Arg-Leu-Leu-Pro-His (RLLPH), derived from the C3 fraction, attenuated adipogenesis by downregulating the expression of several mRNAs related to adipogenesis, including peroxisome prolif...

Partition efficiency of cytochrome c with alcohol/salt aqueous biphasic flotation system
Publication date: Available online 17 October 2019Source: Journal of Bioscience and BioengineeringAuthor(s): Hui-Suan Ng, Tze-Cheng Ng, Phei Er Kee, Joo Shun Tan, Hip Seng Yim, John Chi-Wei LanAqueous biphasic flotation (ABF) integrates aqueous biphasic system (ABS) and solvent sublation for recovery of target biomolecules. The feasibility of the alcohol/salt ABF for exclusive partition of cytochrome c to one specific phase of the system was investigated. Aliphatic alcohols of different carbon chain length (ethanol, 1-propanol and 2-propanol) and salts (sulfate, phosphate and citrate) were used for the phase formation. The...
