CXCL12-CXCR4 mediates CD57 < sup > + < /sup > CD8 < sup > + < /sup > T cell responses in the progression of type 1 diabetes
J Autoimmun. 2024 Jan 31;143:103171. doi: 10.1016/j.jaut.2024.103171. Online ahead of print.ABSTRACTCD57+ CD8+ T cells, also referred as effector memory cells, are implicated in various conditions including tumor immunity, virus immunity, and most recently with autoimmunity. However, their roles in the progression and remission of T1D are still unclear. Here, we noted an increase in peripheral CD57+ CD8+ T cells in a T1D patient harboring an activator of transcription 3 (STAT3) mutation. Our in-depth study on the role of CD57+ CD8+ T cells within a T1D patient cohort revealed that these cells undergo significant compositio...

IL-23 induces CLEC5A < sup > + < /sup > IL-17A < sup > + < /sup > neutrophils and elicit skin inflammation associated with psoriatic arthritis
J Autoimmun. 2024 Jan 31;143:103167. doi: 10.1016/j.jaut.2024.103167. Online ahead of print.ABSTRACTIL-23-activation of IL-17 producing T cells is involved in many rheumatic diseases. Herein, we investigate the role of IL-23 in the activation of myeloid cell subsets that contribute to skin inflammation in mice and man. IL-23 gene transfer in WT, IL-23RGFP reporter mice and subsequent analysis with spectral cytometry show that IL-23 regulates early innate immune events by inducing the expansion of a myeloid MDL1+CD11b+Ly6G+ population that dictates epidermal hyperplasia, acanthosis, and parakeratosis; hallmark pathologic fe...

Immunosuppression induces regression of fibrosis in primary biliary cholangitis with moderate-to-severe interface hepatitis
CONCLUSION: Combination therapy with IS improves outcomes in PBC patients with moderate-to-severe interface hepatitis compared to UDCA monotherapy. Baseline IgG and ALP are the most significant predictors of fibrosis regression. The new predictive score, FRS, incorporating baseline IgG and ALP, can effectively identify individuals who would benefit from the combination therapy.PMID:38301505 | DOI:10.1016/j.jaut.2023.103163 (Source: Journal of Autoimmunity)

Older age of celiac disease diagnosis and risk of autoimmune disease: A nationwide matched case-control study
CONCLUSIONS: This study revealed an increased risk of autoimmune disease among CeD patients compared with controls. However, older age at CeD diagnosis did not seem to escalate the risk of autoimmune diseases.PMID:38286066 | DOI:10.1016/j.jaut.2024.103170 (Source: Journal of Autoimmunity)

A survey of ficolin-3 activity in Systemic Lupus Erythematosus reveals a link to hematological disease manifestations and autoantibody profile
J Autoimmun. 2024 Jan 13;143:103166. doi: 10.1016/j.jaut.2023.103166. Online ahead of print.ABSTRACTThe complement system plays a central role in the pathogenesis of Systemic Lupus Erythematosus (SLE), but most studies have focused on the classical pathway. Ficolin-3 is the main initiator of the lectin pathway of complement in humans, but its role in systemic autoimmune disease has not been conclusively determined. Here, we combined biochemical and genetic approaches to assess the contribution of ficolin-3 to SLE risk and disease manifestations. Ficolin-3 activity was measured by a functional assay in serum or plasma sampl...

Proximity extension assay proteomics and renal single cell transcriptomics uncover novel urinary biomarkers for active lupus nephritis
CONCLUSION: In addition to confirming the diagnostic potential of urine ALCAM, CD163, MCP1, SELL, ICAM1, VCAM1, NGAL and TWEAK for active LN, this study adds urine ICAM-2, FABP4, FASLG, IGFBP-2, SELE, and TNFSF13B/BAFF as additional markers that warrant systematic validation in larger cross-sectional and longitudinal cohorts.PMID:38194790 | DOI:10.1016/j.jaut.2023.103165 (Source: Journal of Autoimmunity)

Single-cell sequencing of the retina shows that LDHA regulates pathogenesis of autoimmune uveitis
In this study, single-cell RNA sequencing of the retina and cervical draining lymph nodes in experimental autoimmune uveitis mice was conducted to identify the cellular spatiotemporal dynamics and upregulation of the glycolysis-related gene LDHA. Suppression of LDHA can rescue the imbalance of T effector (Teff) cells/T regulator (Treg) cells under inflammation via downregulation of the glycolysis-PI3K signaling circuit and inhibition of the migration of CXCR4+ Teff cells towards retinal tissue. Furthermore, LDHA and CXCR4 are upregulated in the peripheral blood mononuclear cells of Vogt-Koyanagi-Harada patients. The LDHA i...

Silencing of aryl hydrocarbon receptor repressor restrains Th17  cell immunity in autoimmune hepatitis
In conclusion, blockade of AHRR in AIH restores Th17-cell response to AHR, and limits Th17-cell differentiation through generation of uridine. In vivo, silencing of AHRR attenuates liver damage in NOD/scid/gamma mice. Blockade of AHRR might therefore represent a novel therapeutic strategy to modulate effector Th17-cell immunity and restore homeostasis in AIH.PMID:38142533 | DOI:10.1016/j.jaut.2023.103162 (Source: Journal of Autoimmunity)
