Mitochondrial DNA mutations in HIV-exposed uninfected infants following the cessation of triple antiretroviral drugs
ConclusionsFurther studies are needed to determine the effects of these mutations on the development of HEU infants and whether stopping ART leads to the restoration of mitochondrial function. (Source: Journal of Antimicrobial Chemotherapy)

Effect of albumin substitution on pharmacokinetics of piperacillin/tazobactam in patients with severe burn injury admitted to the ICU
ConclusionsAlbumin supplementation had little impact on the PK of piperacillin/tazobactam. After albumin supplementation, there was a numerical increase in mean AUC0 –8 of total and unbound piperacillin/tazobactam, whereas similarCmax values were observed. Future studies may investigate the effect of albumin supplementation on drugs with a higher plasma protein binding. (Source: Journal of Antimicrobial Chemotherapy)

Efficacy and safety of raltegravir plus lamivudine maintenance therapy
ConclusionsSwitching to raltegravir and lamivudine in PWH with virological suppression maintains efficacy and is well tolerated. This maintenance regimen might be a cost-effective option for PWH at risk of drug –drug interactions or needing to avoid specific toxicities of certain antiretroviral drugs or their negative impact on comorbidities. Imipenem/funobactam (formerly XNW4107) in vivo pharmacodynamics against serine carbapenemase-producing Gram-negative bacteria: a novel modelling approach for time-dependent killing
ConclusionsImipenem/funobactam showed potentin vivo efficacy against serine carbapenemase-producers. The novel PK/PD index (%fT  >  CT)/MIC appeared to best describein vivo activity. (Source: Journal of Antimicrobial Chemotherapy)

Novel SCCmec variants in clonal complex 398 and lineage-specific pseudo-SCCmec identified in ST88 MRSA from invasive bloodstream infections in China
ConclusionsOur findings provide evidence for the ongoing evolution of SCCmec elements within the CC398 and ST88 clones, underscoring the need for further surveillance to understand the biological significance of these elements. (Source: Journal of Antimicrobial Chemotherapy)

Surveillance of antimicrobial resistant Shiga toxin-producing E. coli O157:H7 in England, 2016 –2020
ConclusionsAMR genes were unevenly distributed across the different sub-lineages of STEC O157:H7 and between different clades within the same sub-lineage. Long-read sequencing facilitates tracking the transmission of AMR at the pathogen and mobile genetic element level. (Source: Journal of Antimicrobial Chemotherapy)

Prevalence of HIV-1 drug resistance mutations in proviral DNA in the Swiss HIV Cohort Study, a retrospective study from 1995 to 2018
ConclusionsWe successfully applied a reliable and efficient method to analyse near full-length HIV-1 proviral DNA and investigated DRMs in individuals with undetectable or low viraemia. Additionally, our data underscore the need for new computational tools to exclude APOBEC-related hypermutated NGS sequence reads for reporting DRMs. (Source: Journal of Antimicrobial Chemotherapy)

Decay kinetics of HIV-1-RNA in seminal plasma with dolutegravir/lamivudine versus dolutegravir plus emtricitabine/tenofovir alafenamide in treatment-naive people living with HIV
ConclusionsAfter 12 weeks of treatment, there were no differences in the percentage of undetectable SP HIV-1-RNA in naive PLWH who started dolutegravir/lamivudine compared with DTG  + TAF/FTC. (Source: Journal of Antimicrobial Chemotherapy)

Effect of dapagliflozin on COVID-19 infection and risk of hospitalization
ConclusionsUse of dapagliflozin prior to SARS-CoV-2 infection was not associated with an increased risk of hospitalization, ICU admission, mortality or progression to severe COVID-19. However, it was associated with an increased risk of susceptibility to COVID-19 infection. (Source: Journal of Antimicrobial Chemotherapy)

Long-term effects on subclinical cardiovascular disease of switching from boosted protease inhibitors to dolutegravir
DiscussionAfter 96 weeks, switching from PI/r to dolutegravir in PWH with high cardiovascular risk led to significant changes in several biomarkers associated with cardiovascular disease. Although most changes were favourable, adiponectin reduction was not. There were non-significant changes in CIMT progression. (Source: Journal of Antimicrobial Chemotherapy)

Significant increase of CTX-M-15-ST131 and emergence of CTX-M-27-ST131 Escherichia coli high-risk clones causing healthcare-associated bacteraemia of urinary origin in Spain (ITUBRAS-2 project)
ConclusionsST131 HiRC is the main driver of ESBL-producingE. coli causing HCA-BUO in Spain, mainly associated with the expansion of subclade CTX-M-15-C2/H30-Rx and the emergence of CTX-M-27-C1/H30-R1 (Cluster C1-M27). (Source: Journal of Antimicrobial Chemotherapy)

Blood telomere length gain in people living with HIV switching to dolutegravir plus lamivudine versus continuing triple regimen: a longitudinal, prospective, matched, controlled study
ConclusionsIn this setting of virologically suppressed PLWH, simplifying to dolutegravir  + lamivudine was associated with a higher gain in BTL than maintaining triple therapy after the 1 year follow-up. These findings suggest that as a simplification strategy dolutegravir + lamivudine might have a positive effect on BTL. (Source: Journal of Antimicrobial Chemotherapy)

The Neonatal and Paediatric Pharmacokinetics of Antimicrobials study (NAPPA): investigating amoxicillin, benzylpenicillin, flucloxacillin and piperacillin pharmacokinetics from birth to adolescence
ConclusionsNAPPA represents, to our knowledge, the largest prospective combined paediatric penicillin PK study undertaken to date, and the first paediatric flucloxacillin oral PK model. The PTA results provide evidence supportive of BNFC high-dose IV regimens for amoxicillin, benzylpenicillin and piperacillin. (Source: Journal of Antimicrobial Chemotherapy)

Antibacterial effect of seven days exposure to ceftolozane-tazobactam as monotherapy and in combination with fosfomycin or tobramycin against Pseudomonas aeruginosa with ceftolozane-tazobactam MICs at or above 4 mg/l in an in vitro pharmacokinetic model
ConclusionsThe addition of either fosfomycin or tobramycin to ceftolozane/tazobactam at simulated human clinically observed concentrations reducedP. aeruginosa bacterial loads and the risk of resistance to ceftolozane/tazobactam when strains had ceftolozane/tazobactam MIC values at or above the clinical breakpoint.
