Effect of a personalized intensive dietary intervention on Base Excision Repair (BER) in colorectal cancer patients: Results from a randomized controlled trial
In conclusion, the results do not support an effect of dietary intervention on BER activity in post-surgery CRC patients during a 6-month intervention period.PMID:38588903 | DOI:10.1016/j.freeradbiomed.2024.04.211 (Source: Free Radical Biology and Medicine)

Activation of lipophagy is required for RAB7 to regulate ferroptosis in sepsis-induced acute kidney injury
This study investigated the mechanism by which lipid droplets (LDs) degraded via autophagy (lipophagy)required for RAB7 regulated ferroptosis in the pathogenesis of S-AKI. Here, we constructed the S-AKI model in vitro and in vivo to elucidate the potential relationship of lipophagy and ferroptosis, and we first confirmed that the activation of lipophagy promoted renal tubular epithelial cell ferroptosis and renal damage in S-AKI. The results showed that lipopolysaccharide (LPS) induced a marked increase in lipid peroxidation and ferroptosis, which were rescued by ferrstain-1 (Fer-1), an inhibitor of ferroptosis. In additio...

Doxorubicin induces phosphorylation of lamin A/C and loss of nuclear membrane integrity: A novel mechanism of cardiotoxicity
Free Radic Biol Med. 2024 Apr 4:S0891-5849(24)00383-6. doi: 10.1016/j.freeradbiomed.2024.04.212. Online ahead of print.ABSTRACTLamin A/C, essential inner nuclear membrane proteins, have been linked to progeria, a disease of accelerated aging, and many other diseases, which include cardiac disorder. Lamin A/C mutation and its phosphorylation are associated with altering nuclear shape and size. The role of lamin A/C in regulating normal cardiac function was reported earlier. In the present study, we hypothesized that Doxorubicin (Dox) may alter total lamin A/C expression and phosphorylation, thereby taking part in cardiac in...

TET3 boosts hepatocyte autophagy and impairs non-alcoholic fatty liver disease by increasing ENPP1 promoter hypomethylation
CONCLUSIONS: We describe a novel epigenetic mechanism wherein TET3 promoted ENPP1 expression through promoter hypomethylation is a critical mediator of NAFLD. Our findings provide new insight into the development of preventative measures for NAFLD.PMID:38582229 | DOI:10.1016/j.freeradbiomed.2024.04.207 (Source: Free Radical Biology and Medicine)

Peroxiredoxin 6 suppresses ferroptosis in lung endothelial cells
Free Radic Biol Med. 2024 Apr 3:S0891-5849(24)00373-3. doi: 10.1016/j.freeradbiomed.2024.04.208. Online ahead of print.ABSTRACTPeroxiredoxin 6 (Prdx6) repairs peroxidized membranes by reducing oxidized phospholipids, and by replacing oxidized sn-2 fatty acyl groups through hydrolysis/reacylation by its phospholipase A2 (aiPLA2) and lysophosphatidylcholine acyltransferase activities. Prdx6 is highly expressed in the lung, and intact lungs and cells null for Prdx6 or with single-point mutations that inactivate either Prdx6-peroxidase or aiPLA2 activity alone exhibit decreased viability, increased lipid peroxidation, and inco...

Mitochondrial quality control in non-exudative age-related macular degeneration: From molecular mechanisms to structural and functional recovery
Free Radic Biol Med. 2024 Apr 3:S0891-5849(24)00161-8. doi: 10.1016/j.freeradbiomed.2024.03.024. Online ahead of print.ABSTRACTNon-exudative age-related macular degeneration (NE-AMD) is the leading blindness cause in the elderly. Clinical and experimental evidence supports that early alterations in macular retinal pigment epithelium (RPE) mitochondria play a key role in NE-AMD-induced damage. Mitochondrial dynamics (biogenesis, fusion, fission, and mitophagy), which is under the central control of AMP-activated kinase (AMPK), in turn, determines mitochondrial quality. We have developed a NE-AMD model in C57BL/6J mice induc...
