Reduced Zn < sup > 2+ < /sup > promotes retinal ganglion cells survival and optic nerve regeneration after injury through inhibiting autophagy mediated by ROS/Nrf2
This study verified that TPEN or knocking out ZnT-3 in ACs is a promising therapeutic option for the treatment of optic nerve damage and elucidated the postsynaptic molecular mechanism of Zn2+-triggered damage to RGCs after ONC injury.PMID:38134974 | DOI:10.1016/j.freeradbiomed.2023.12.008 (Source: Free Radical Biology and Medicine)

Oxidation of the active site cysteine residue of glyceraldehyde-3-phosphate dehydrogenase to the hyper-oxidized sulfonic acid form is favored under crowded conditions
In this study, we investigated the role of crowding (induced by dextran) on H2O2- and SIN-1-induced GAPDH oxidation, with data for crowded and dilute conditions compared. LC-MS/MS data revealed a lower extent of modification of the catalytic Cys under crowded conditions (i.e. less monomer units modified), but enhanced formation of the sulfonic acid resulting from hyper-oxidation. This effect was not observed with SIN-1. These data indicate that molecular crowding can modulate the oxidation pathways of GAPDH and its extent of oxidation and inactivation.PMID:38122871 | DOI:10.1016/j.freeradbiomed.2023.12.015 (Source: Free Ra...

Evaluation of a novel pyridinium cation-linked styryl-based boronate probe for the detection of selected inflammation-related oxidants
Free Radic Biol Med. 2023 Dec 18:S0891-5849(23)01148-6. doi: 10.1016/j.freeradbiomed.2023.12.003. Online ahead of print.ABSTRACTReactive oxygen and nitrogen species (RONS) are a range of chemical individuals produced by living cells that contribute to the proper functioning of organisms. Cells under oxidative and nitrative stress show excessive production of RONS (including hydrogen peroxide, H2O2, hypochlorous acid, HOCl, and peroxynitrite, ONOO-) which may result in a damage proteins, lipids, and genetic material. Thus, the development of probes for in vivo detection of such oxidants is an active area of research, focusi...

ALDH2 deficiency exacerbates MCD-diet induced MASLD by modulating bile acid metabolism
Free Radic Biol Med. 2023 Dec 15:S0891-5849(23)01173-5. doi: 10.1016/j.freeradbiomed.2023.12.017. Online ahead of print.ABSTRACTAldehyde dehydrogenase 2 (ALDH2), an acetaldehyde dehydrogenase in mitochondria, is primarily responsible for metabolizing alcohol-derived acetaldehyde and other endogenous aldehydes. Inactivating ALDH2 rs671 polymorphism is found in up to 8 % of the global population and 40 % of the East Asian population. Recent studies have shown that rs671 SNP mutation in the human ALDH2 gene is associated with an increased risk of metabolic dysfunction-associated steatotic liver disease (MASLD), but the mechan...

TNF- α induced NF-κB mediated LYRM7 expression modulates the tumor growth and metastatic ability in breast cancer
Free Radic Biol Med. 2023 Dec 15:S0891-5849(23)01174-7. doi: 10.1016/j.freeradbiomed.2023.12.018. Online ahead of print.ABSTRACTTumor microenvironment (TME) of solid tumors including breast cancer is complex and contains a distinct cytokine pattern including TNF-α, which determines the progression and metastasis of breast tumors. The metastatic potential of triple negative breast cancer subtypes is high as compared to other subtypes of breast cancer. NF-κB is key a transcription factor regulating inflammation and mitochondrial bioenergetics including oxidative phosphorylation (OXPHOS) genes which determine its oxidative ...

Impact of amino acid substitutions in hepatitis C virus core region on the severe oxidative stress
CONCLUSIONS: HCV core proteins harboring either or both substitutions of R70Q/H or L91 M enhanced hepatocellular oxidative stress in vivo and in vitro. These amino acid substitutions may affect HCC development by enhancing hepatic oxidative stress in patients with chronic HCV-1b infection.PMID:38103659 | DOI:10.1016/j.freeradbiomed.2023.12.014 (Source: Free Radical Biology and Medicine)

Bisphenol A induces placental ferroptosis and fetal growth restriction via the YAP/TAZ-ferritinophagy axis
Free Radic Biol Med. 2023 Dec 14:S0891-5849(23)01158-9. doi: 10.1016/j.freeradbiomed.2023.12.013. Online ahead of print.ABSTRACTExposure to bisphenol A (BPA) during gestation leads to fetal growth restriction (FGR), whereby the underlying mechanisms remain unknown. Here, we found that FGR patients showed higher levels of BPA in the urine, serum, and placenta; meanwhile, trophoblast ferroptosis was observed in FGR placentas, as indicated by accumulated intracellular iron, impaired antioxidant molecules, and increased lipid peroxidation products. To investigate the role of ferroptosis in placental and fetal growth, BPA stimu...
