Trimethylamine N-oxide promotes abdominal aortic aneurysm by inducing vascular inflammation and vascular smooth muscle cell phenotypic switching
CONCLUSIONS: TMAO promotes AAA formation by inducing vascular inflammation and VSMC phenotypic switching through activation of the NF-κB signaling pathway. Thus, TMAO is a prospective therapeutic AAA target.PMID:38160930 | DOI:10.1016/j.ejphar.2023.176307 (Source: European Journal of Pharmacology)

Mechanism of GW117 antidepressant action: Melatonin receptor-mediated regulation of sleep rhythm
Eur J Pharmacol. 2023 Dec 29:176299. doi: 10.1016/j.ejphar.2023.176299. Online ahead of print.ABSTRACTAlterations in circadian sleep patterns constitute a salient manifestation in major depressive disorder. GW117, an emergent antidepressant, functions as an agonist for melatonin 1 and melatonin 2 (MT1/MT2) receptors, in tandem with antagonism of the serotonin (5-HT) 2C receptor. The present investigation is dedicated to elucidating the role and underlying mechanisms by which GW117 ameliorates circadian sleep disruptions. Utilizing an adapted chronic unpredictable mild stress protocol, we induced a depressive-like phenotype...

Andrographolide derivative Andro-III modulates neuroinflammation and attenuates neuropathological changes of Alzheimer's disease via GSK-3 β/NF-κB/CREB pathway
This study aims to examine the impact of Andro-III on enhancing cognitive learning ability in 3xTg-AD mice, as well as the mechanisms involved. Andro-III improved spatial learning ability, prevented the loss of Nysted's vesicles, reduced the accumulation of β-amyloid (Aβ) and tau proteins, and suppressed microglial activation. Further research found that the expression of nuclear factor kappa-B RelA (NF-κB p65) expression and glycogen synthase kinase-3β (GSK-3β) activity were inhibited, while CREB was upregulated in brain tissue treated with Andro-III. Moreover, Andro-III downregulated the expression of IBA1 and infla...

Nur77 alleviates cardiac fibrosis by upregulating GSK-3 β transcription during aging
This study proposes a novel mechanism for Nur77 regulating cardiac fibrosis and suggests Nur77 as a target for the prevention and treatment of aging-associated cardiac fibrosis and heart failure.PMID:38158109 | DOI:10.1016/j.ejphar.2023.176290 (Source: European Journal of Pharmacology)

Complanatuside A improves functional recovery after spinal cord injury through inhibiting JNK signaling-mediated microglial activation
CONCLUSIONS: ComA can accelerate the restoration of motor function in mice after SCI, possibly by reducing neuronal apoptosis via inhibition of JNK-related signaling pathways, a reduction in microglial activation, and inhibition of inflammatory factor release. Our data indicate that ComA is a promising drug candidate for improving functional recovery in patients with SCI.PMID:38158110 | DOI:10.1016/j.ejphar.2023.176287 (Source: European Journal of Pharmacology)
