Enzymatic degradation pattern of polysorbate 20 impacts interfacial properties of monoclonal antibody formulations
In this study, two hydrolytically-active immobilized lipases, which primarily targeted either monoester or higher-order ester species in PS20, were used to generate partially-degraded PS20. The impact of PS20 degradation pattern on critical micelle concentration (CMC), surface tension, interfacial rheology parameters and agitation protection was assessed. CMC was slightly increased upon monoester degradation, but significantly increased upon higher-order esters degradation. The PS20 degradation pattern also significantly impacted the dynamic surface tension of a mAb formulation, whereas changes in the equilibrium surface t...

Combination Techniques Towards Novel Drug Delivery Systems Manufacturing: 3D PCL Scaffolds Enriched with Tetracycline-loaded PVP Nanoparticles
Eur J Pharm Biopharm. 2023 Nov 28:S0939-6411(23)00311-9. doi: 10.1016/j.ejpb.2023.11.022. Online ahead of print.ABSTRACTDrug delivery systems based on synthetic and natural polymers offer a new approach with a capacity to control the release of bio-active agents within time. In this work, we present different designs of Polycaprolactone (PCL) 3D scaffolds containing Polyvinylpyrrolidone (PVP) nanoparticles that can store a hydrophilic drug. The drug delivery system, combined of PCL and PVP polymers fabricated by additive manufacturing, aims for a solution for longer and more stabled drug delivery carrier. The drug, planned...

Influence of water and trehalose on α- and β-relaxation of freeze-dried lysozyme formulations
Eur J Pharm Biopharm. 2023 Nov 27:S0939-6411(23)00308-9. doi: 10.1016/j.ejpb.2023.11.019. Online ahead of print.ABSTRACTMolecular mobility in form of alpha and beta relaxations is considered crucial for characterization of amorphous lyophilizates and reflected in the transition temperatures Tgα and Tgβ. Based on an overview of applied methods to study beta relaxations, Dynamic Mechanical analysis was used to measure Tgα and Tgβ in amorphous freeze-dried samples. Lysozyme and trehalose as well as their mixtures in varying ratios were investigated. Three different residual moisture levels, ranging from roughly 0.5-7 % (w...

Rapid liposomal formulation for nucleolin targeting to rhabdomyosarcoma cells
Eur J Pharm Biopharm. 2023 Nov 27:S0939-6411(23)00309-0. doi: 10.1016/j.ejpb.2023.11.020. Online ahead of print.ABSTRACTRhabdomyosarcoma (RMS) is the most common pediatric soft tissue sarcoma. More effective and less toxic therapies are urgently needed for high-risk patients. Peptide-guided targeted drug delivery can increase the therapeutic index of encapsulated drugs and improve patients' well-being. To apply this strategy to RMS, we identified the peptide F3 in a screening for peptides binding to RMS cells surface. F3 binds to nucleolin, which is present on the surface of RMS cells and is abundantly expressed at the mRN...
